Numerical investigation of deposition mechanism in three mouth-throat models

F. Huang et al.

Summary: This study investigated the effects of geometrical variation, particle size, and airflow rate on fine particle deposition in the mouth-throat. The results showed that geometrical variation and respiratory conditions significantly influenced both deposition fraction and spatial distribution, with geometrical variation playing a more dominant role in particle escape pattern.

Role of CFD based in silico modelling in establishing an in vitro-in vivo correlation of aerosol deposition in the respiratory tract

Fen Huang et al.

Summary: The effective evaluation and prediction of aerosol transport deposition in human respiratory tracts are critical to aerosol drug delivery and inhalation product evaluation. Computational fluid dynamics (CFD) based in silico modeling has rapidly developed in the past two decades, particularly with the coupling of discrete phase model (DPM) and discrete element method (DEM) approaches. Future research recommendations include improving in silico prediction to achieve better in vitro -in vivo correlation (IVIVC).

Estimating inter-patient variability of dispersion in dry powder inhalers using CFD-DEM simulations

Benedict Benque et al.

Summary: Drug delivery from a capsule-based dry powder inhaler is influenced by the inhaler's design, the drug's formulation, and the inhalation maneuver. Patient variability poses a challenge in designing new inhaler types. Simulation studies show that different inhalation profiles can affect carrier powder motion and acceleration events.

Porous particles and novel carrier particles with enhanced penetration for efficient pulmonary delivery of antitubercular drugs

Jun Yee Tse et al.

Summary: This study aimed to enhance the pulmonary delivery of drugs by using PyG as a multi-functional additive to increase macrophage-like cell phagocytic activity. The dry powder inhaler formulations of rifampicin with PyG showed improved inhalation performance, with PyG facilitating cellular uptake and inflammatory cytokine gene expression. The inclusion of PyG in porous and wrinkled particle formulations significantly enhanced their aerosolization properties and may advance research in improving the therapeutic effect of antitubercular drugs.

Incorrect Holding Angle of Dry Powder Inhaler during the Drug-Loading Step Significantly Decreases Output Efficiency

Kana Suenaga et al.

Summary: The study revealed that 33.3% of participants held the inhaler at an incorrect angle during the drug-loading step. In vitro experiments showed that holding the inhaler at an incorrect angle significantly decreased OE (an index of the loaded dose) by 73.3%, while St2 (an index of the deagglomerating efficiency) remained stable regardless of the holding angle. OE x St2, indicating the bronchial and pulmonary drug delivery amount, decreased by 76.9%. Holding the inhaler at an incorrect angle reduces the loaded dose, leading to decreased pulmonary delivery.

Flow Structure and Particle Deposition Analyses for Optimization of a Pressurized Metered Dose Inhaler (pMDI) in a Model of Tracheobronchial Airway

Kaveh Ahookhosh et al.

Summary: Inhalation therapy is crucial for the management of respiratory diseases, and pressurized metered dose inhalers (pMDIs) are a popular drug delivery device. Computational study on flow structure inside pMDI and deposition of micron-sized particles in human airway model revealed significant impact of inhalation air flow rate and inhaler geometry on deposition patterns.
