Physicochemical properties of nanoparticles affecting their fate and the physiological function of pulmonary surfactants

Summary: This review summarizes how the physicochemical properties of nanoparticles affect the physiological function and fate of pulmonary surfactants. The composition and characteristics of pulmonary surfactants, as well as the methods for studying their interaction with nanoparticles, are introduced. The influence of nanoparticles' physicochemical properties on hydrophobic protein adsorption and strategies to decrease their interaction with pulmonary surfactants are discussed. The influence of nanoparticles' physicochemical properties on lipids and hydrophilic protein adsorption and their consequent fate is described.

Inhalable PLGA microspheres: Tunable lung retention and systemic exposure via polyethylene glycol modification

Summary: Polyethylene glycol (PEG) modification is a promising approach to overcome barriers in the deep lung for sustained therapy of pulmonary diseases. PEG-modified microspheres showed enhanced mucus permeability and reduced macrophage uptake, especially the PEG(2000)-DSPE/PLGA 1:1 group. The PEG 2000-based group had significantly prolonged lung retention in vivo and reduced systemic exposure of drugs compared to other groups, making it a promising candidate for sustained lung delivery in pulmonary disease treatment.

Summary: By investigating the interactions between nanoparticles and pulmonary surfactant, researchers found that surfactant preparations with high interfacial activity and distinctive composition can reduce the toxicity of aerosolized particles and help decrease the aggregation of aerosolized particles. These findings help identify key requirements when studying interactions of nanomaterials with the pulmonary air-blood barrier in vitro.
