Cationic Antimicrobial Peptides Are Leading the Way to Combat Oropathogenic Infections

Summary: Oral dental infections, primarily caused by oral bacterial pathogens, are common in humans with caries and periodontal diseases being the most prevalent. Misuse of antibiotics has led to antimicrobial resistance, but studies show that cationic antimicrobial peptides are effective against oral pathogens with less propensity for resistance development.

An optimized analog of antimicrobial peptide Jelleine-1 shows enhanced antimicrobial activity against multidrug resistant P. aeruginosa and negligible toxicity in vitro and in vivo

Summary: Novel analogs of jelleine-1 were designed, synthesized, and examined for their antimicrobial functions and toxicity, showing improved bioactivity through amino acid substitutions at specific positions. The substitution of phenylalanine by tryptophan also enhanced the antimicrobial activity. Among all the analogs, analog 15 enriched in arginine and leucine exhibited the most potent antimicrobial activity against both gram-negative and gram-positive bacteria, especially multi-drug resistant P. aeruginosa, with potent inhibition of biofilm formation and negligible toxicity.
