Targeted cancer treatment and fertility: effect of immunotherapy and small molecule inhibitors on female reproduction

Parker L. Bussies et al.

Summary: Targeted cancer therapy plays an important role in improving survival outcomes for reproductive aged patients, but its effects on the female reproductive system are not well understood. Data-driven recommendations are needed to help clinicians counsel women on fertility issues.

Malignant melanoma treated with pembrolizumab during pregnancy: A case report and review of the literature

Yuki Anami et al.

Summary: There is limited research on using immune checkpoint inhibitors for malignant tumors during pregnancy. This study presents a case of a 40-year-old primigravida with advanced malignant melanoma who was treated with pembrolizumab during pregnancy and successfully gave birth with controlled disease.

Risks of molecular targeted therapies to fertility and safety during pregnancy: a review of current knowledge and future needs

Elena Lorenzi et al.

Summary: As the population of young cancer survivors increases and a trend towards postponing pregnancy later in life is reported, more efforts are focused on understanding treatment-induced sequelae, especially the effects of cancer and/or treatment on fertility. While most MTTs have worrying preclinical data discouraging their use during pregnancy, it does not necessarily mean that all MTTs result in permanent infertility and should be completely avoided during pregnancy. This review provides a critical evaluation on the most commonly used MTTs, offering a possible guide for clinicians.

A retrospective audit of cumulative ionising radiation levels in hospitalised pregnant patients

James H. McEvoy-May et al.

Summary: This study assessed the cumulative levels of ionising radiation received by pregnant patients during a single admission to a tertiary hospital in Australia. The results indicate that pregnant patients are exposed to low doses of ionising radiation, with stays longer than ten days and maternal cardiovascular related admissions associated with significantly higher cumulative effective doses for both mothers and fetuses.

A review of the types of childhood cancer associated with a medical X-ray examination of the pregnant mother

Richard Wakeford et al.

Summary: Studies have shown consistent elevations of risk for various types of childhood cancer, including leukemia, with antenatal X-ray exposure, although results vary when considering categories with limited data. The interpretational value of cohort studies is limited due to low statistical power and other identified shortcomings, while the overall combination of other case-control/case-cohort studies supports the inference that low-level radiation exposure in utero proportional increases the risk of childhood cancers.

Adjuvant pembrolizumab versus placebo in resected stage III melanoma (EORTC 1325-MG/KEYNOTE-054): distant metastasis-free survival results from a double-blind, randomised, controlled, phase 3 trial

Alexander M. M. Eggermont et al.

Summary: The study demonstrated that adjuvant pembrolizumab therapy can improve the 3.5-year distant metastasis-free survival in patients with resected high-risk stage III cutaneous melanoma, with a more significant effect in patients with PD-L1-positive tumors.

Estrogen Receptor Is Expressed in Uveal Melanoma: A Potential Target for Therapy

Lynn Schoenfield et al.

Summary: Immunohistochemical analysis showed that over half of UM cases express ER. Additionally, elevated gene expression of ESR1 and ESR2 was identified in a subset of UM tumors with poor genetic prognostic features. These findings support the potential targeting of ER for therapy in UM.

Summary: The review highlights the potential impact of pregnancy on uveal melanoma development and progression, as well as the lack of significant effect from oral contraceptives and hormone replacement therapy on incidence. Current treatment strategies for pregnant UM patients mainly focus on targeted receptor-based chemotherapy.
