The matrix metalloproteinase inhibitor IPR-179 has antiseizure and antiepileptogenic effects

Diede W. M. Broekaart et al.

Summary: Research has shown that the expression of MMPs and TIMPs is persistently dysregulated in the hippocampus of patients with epilepsy and experimental animals. Treatment with the MMP2/9 inhibitor IPR-179 can reduce seizure severity and improve cognitive behavior in rodent models of epilepsy. It also prevents an MMP2/9-dependent switch-off that normally restrains network excitability, suggesting potential for further investigation in clinical trials.

Low prevalence of amyloid and tau pathology in drug-resistant temporal lobe epilepsy

Juliana C. Silva et al.

Summary: A beta plaques and pTau were not commonly found in resected brain tissue of temporal lobe epilepsy patients undergoing surgery, and did not show significant correlation with clinical characteristics or preoperative psychometric test scores, except for a lower median score on the VPA hard assessment in patients with A beta plaques. Therefore, cognitive impairment in TLE is less likely to be driven by the same mechanisms as in Alzheimer disease.

Seizures are a druggable mechanistic link between TBI and subsequent tauopathy

Hadeel Alyenbaawi et al.

Summary: Research suggests that traumatic brain injury (TBI) may contribute to tauopathy and subsequent dementia, with seizures potentially playing a key role in promoting the spread of Tau aggregates. Using a zebrafish model, it was found that bath application of dynamin inhibitors or anticonvulsant drugs could rescue TBI-induced tauopathy and cell death. Further exploration is needed to investigate the potential of anti-convulsants in mitigating post-traumatic seizures and tauopathy.

Peripheral Blood and Salivary Biomarkers of Blood-Brain Barrier Permeability and Neuronal Damage: Clinical and Applied Concepts

Damir Janigro et al.

Summary: The blood-brain barrier in the neurovascular unit plays a critical role in isolating the brain parenchyma from peripheral blood, and increased permeability of the barrier can contribute to the onset of CNS diseases. Biomarkers from glial, neuronal, and pericyte cells can exit the brain and appear in peripheral body fluids, such as saliva or urine, in various temporal trajectories under pathological conditions.

Neurovascular multiparametric MRI defines epileptogenic and seizure propagation regions in experimental mesiotemporal lobe epilepsy

Fabien Boux et al.

Summary: The study aims to improve the identification of the epileptogenic zone and associated seizure-spreading regions using multi-parametric MRI analysis. The results show that combining multiple cellular and cerebrovascular MRI read-outs can effectively differentiate epileptogenic from seizure-spreading areas in an experimental model of mesial temporal lobe epilepsy. The potential clinical value of these findings is discussed.

Magnetic Resonance Imaging of Blood-Brain Barrier permeability in Dementia

Audrey Chagnot et al.

Summary: Alzheimer's disease and cerebral small vessel disease are two major causes of dementia, with blood-brain barrier breakdown being a common factor. Advances in neuroimaging techniques, such as dynamic contrast-enhanced MRI, allow for the detection of subtle changes in BBB integrity. This review focuses on recent DCEMRI findings related to cSVD and AD, highlighting challenges and pitfalls faced in the context of dementia.

Blood-Brain Barrier Breakdown: An Emerging Biomarker of Cognitive Impairment in Normal Aging and Dementia

Basharat Hussain et al.

Summary: The review discusses the crucial role of the blood-brain barrier in maintaining the specialized microenvironment of neural tissue and the associated BBB breakdown with normal aging and various neurodegenerative diseases. It also covers dementia such as AD and VaD, and the phenotypes and mechanisms of BBB disruption correlated with cognitive decline. The study suggests that BBB breakdown could serve as an emerging biomarker to assist in diagnosing cognitive impairment associated with normal aging and dementia.

Summary: Blood-brain barrier dysfunction is a key factor in cerebral small vessel disease. This highlights the important role of BBB disruption in the pathophysiology and clinical progression of cSVD.
