Effect of Chelation Therapy on a Korean Patient With Brain Manganese Deposition Resulting From a Compound Heterozygous Mutation in the SLC39A14 Gene

Jae-Hyeok Lee et al.

Summary: This study reports the first identified case of SLC39A14-associated manganism in Korea and the response to a 5-year chelation therapy. The treatment successfully reduced serum manganese levels and imaging abnormalities, but the dystonia symptoms showed insignificant improvement. Early diagnosis and treatment are crucial for this genetic disorder.

Molecular Targets of Manganese-Induced Neurotoxicity: A Five-Year Update

Alexey A. Tinkov et al.

Summary: Recent research has uncovered various mechanisms of Mn-induced neurotoxicity, including the role of Mn transporters, metabolic pathways, autophagic flux, synaptic dysfunction, and neuroinflammation, among others. Further studies are needed to fully understand the critical targets and pathways involved in Mn-induced neurotoxicity.

Protein S-nitrosylation and oxidation contribute to protein misfolding in neurodegeneration

Tomohiro Nakamura et al.

Summary: Neurodegenerative disorders such as Alzheimer's and Parkinson's diseases are characterized by progressive degeneration of synapses and neurons, often attributed to accumulated misfolded/aggregated proteins. Excessive reactive oxygen and nitrogen species in the brain may contribute to protein misfolding, potentially exacerbated by genetic mutations and environmental factors. Understanding the role of reactive nitrogen species in post-translational modifications of proteins could lead to therapeutic interventions for neurodegenerative diseases.

Manganese-induced reactive oxygen species activate IκB kinase to upregulate YY1 and impair glutamate transporter EAAT2 function in human astrocytes in vitro

Asha Rizor et al.

Summary: The dysregulation of EAAT2 in neurological disorders, such as Parkinson's disease, Alzheimer's disease, and manganism induced by chronic exposure to high levels of manganese, is associated with NF-kappa B signaling pathway activation and YY1 upregulation. Mn-induced oxidative stress and TNF-alpha production play key roles in the activation of IKK-beta, which leads to the repression of EAAT2 by YY1. Targeting ROS, TNF-alpha, and IKK-beta may be potential strategies to attenuate Mn-induced YY1 activation and subsequent EAAT2 repression.

Reappraisal of metabolic dysfunction in neurodegeneration: Focus on mitochondrial function and calcium signaling

Pooja Jadiya et al.

Summary: The cellular and molecular mechanisms underlying neurodegeneration are still poorly understood, but recent studies have shown that mitochondrial and calcium dysregulation occur early in many NDDs, providing new insights into the pathogenesis. Defective mitochondrial metabolism may play a crucial role in the diverse etiologies of neuropathology.
