A Polycomb repressive complex is required for RNAi-mediated heterochromatin formation and dynamic distribution of nuclear bodies

Jing Xu et al.

Summary: In this study, the EZL1 complex in the protist Tetrahymena thermophila was characterized, revealing its essential role in histone methylation, heterochromatin formation, and genome rearrangement. The interaction between the EZL1 complex and EMA1, as well as PDD1, reinforces its chromatin association and contributes to the dynamic distribution of Polycomb bodies in Tetrahymena development. These findings provide a molecular mechanism linking RNAi and Polycomb repression, ultimately regulating nuclear bodies and genome organization.

Role of Polycomb in the control of transposable elements

Angelique Deleris et al.

Summary: This article summarizes the experimental evidence of Polycomb Repressive Complex 2 (PRC2) targeting and silencing transposable elements across the eukaryotic kingdom, in addition to depositing on silent protein-coding genes, discussing its functional and evolutionary significance. The authors hypothesize that an ancestral role of Polycomb group (PcG) proteins was to silence transposable elements.

Loss of EZH2-like or SU(VAR)3-9-like proteins causes simultaneous perturbations in H3K27 and H3K9 tri-methylation and associated developmental defects in the fungus Podospora anserina

F. Carlier et al.

Summary: Selective gene silencing is crucial for development, with H3K27me3-enriched heterochromatin maintaining transcriptional repression and regulating cell fate. Conversely, H3K9me3-enriched heterochromatin prevents differentiation and protects against transposable elements. Research using the fungus Podospora anserina revealed an interplay between these different heterochromatin conformations, with an interdependence of H3K9me3 and H3K27me3 marks observed. Loss of specific enzymes led to defects in developmental processes, highlighting the importance of histone modifications in fungal development.

Massive colonization of protein-coding exons by selfish genetic elements in Paramecium germline genomes

Diamantis Sellis et al.

Summary: Ciliates are unicellular eukaryotes with a complex genomic architecture including internal eliminated sequences (IESs) that have undergone multiple waves of gains, some of which are under strong purifying selection. The evolution of IESs in Paramecium highlights the important role of selfish genetic elements in shaping genome complexity and gene expression, similar to the evolution of introns in eukaryotes.
