HiCAR is a robust and sensitive method to analyze open-chromatin-associated genome organization

Summary: The long-range interactions of cis-regulatory elements (cREs) are crucial for gene regulation. In this study, we developed a new method called HiCAR (Hi-C on accessible regulatory DNA) to comprehensively identify the spatial interactions of open chromatin sequences. Our results show that HiCAR can efficiently identify high-resolution chromatin contacts in different cell types, with a comparable efficiency to in situ Hi-C. Interestingly, we also discovered that poised gene promoters can silence the expression of distal genes through promoter-promoter interactions. Furthermore, we demonstrated that HiCAR is capable of analyzing chromatin accessibility and looping using low-input primary cells and clinical samples.
