Tissue geometry drives deterministic organoid patterning

N. Gjorevski et al.

Summary: This study describes a method for controlling the formation of epithelial organoids, making the stochastic process more deterministic by spatial and temporal control. By using bioengineering techniques, researchers successfully controlled the formation and patterning of intestinal organoids, and identified the underlying mechanisms of epithelial patterning.

Differentiated Daughter Cells Regulate Stem Cell Proliferation and Fate through Intra-tissue Tension

Wenxiu Ning et al.

Summary: Basal stem cells play a key role in the development, homeostasis, and regeneration of the epidermis, regulated by their microenvironment. Differentiated progeny also influence the proliferation, differentiation, and migration of basal progenitor cells.

Extracellular hyaluronate pressure shaped by cellular tethers drives tissue morphogenesis

Akankshi Munjal et al.

Summary: The formation of zebra fish semicircular canals is primarily driven by hyaluronan secretion instead of conventional actomyosin-driven behaviors. Hyaluronan generates isotropic extracellular pressure to deform the epithelium into buds, while actomyosin and E-cadherin-rich membrane tethers help shape buds into tubes. This study challenges the traditional view that actomyosin contractility is the main driving force in tissue morphogenesis, suggesting that hyaluronate pressure shaped by anisotropic tissue stiffness may be a widespread mechanism for morphological change in organogenesis and tissue engineering.

Adhesion-mediated heterogeneous actin organization governs apoptotic cell extrusion

Anh Phuong Le et al.

Summary: The study reveals that during cell extrusion, the actin network in neighboring cells is reorganized to include lamellipodia protrusions and discontinuous actomyosin cables. The early presence of basal lamellipodia protrusion plays a role in both basal sealing and orienting the actomyosin purse-string, with the coexistence of these mechanisms influenced by cell-cell and cell-substrate adhesions. A theoretical model explains that the integration of these cellular components results in a more efficient extrusion process.

Human neural tube morphogenesis in vitro by geometric constraints

Eyal Karzbrun et al.

Summary: Understanding human organ formation is a challenging scientific task with important medical implications. A chip-based culture system has been developed to recreate neural tube folding from human stem cells, revealing the crucial roles played by neural and non-neural ectoderm in the folding process. The mechanisms of folding involve apical contraction of neural ectoderm and basal adhesion mediated by non-neural ectoderm, with neural tissue width determining neural tube shape. This approach offers a new avenue for studying human organ morphogenesis in both health and disease.

Collective durotaxis along a self-generated stiffness gradient in vivo

Adam Shellard et al.

Summary: The neural crest uses durotaxis to follow stiffness gradients in adjacent placodal tissue, contributing to efficient collective cell migration. Durotaxis synergizes with chemotaxis to polarize actomyosin machinery of the cell group, promoting efficient directional cell migration in vivo.

Retrograde ERK activation waves drive base-to-apex multicellular flow in murine cochlear duct morphogenesis

Mamoru Ishii et al.

Summary: This study explores how the activation waves of extracellular signal-regulated kinase (ERK) control collective cell migration during murine cochlear duct development, promoting duct elongation and development. Through deep tissue live-cell imaging and mathematical modeling, they reveal how the multicellular flow occurring on the lateral side is influenced by the propagating ERK activation.

Inflation-collapse dynamics drive patterning and morphogenesis in intestinal organoids

Naren P. Tallapragada et al.

Summary: This study explores how intestinal organoids self-organize and generate stem cell zones (SCZs) of typical size. The size of SCZs is influenced by ion channel-mediated inflation and mechanosensitive Piezo-family channels, leading to fission through dynamic behavior. Inflation drives stem cell differentiation and induces a stretch-responsive cell state with large transcriptional changes, including upregulation of Piezo1.

Symmetry breaking of tissue mechanics in wound induced hair follicle regeneration of laboratory and spiny mice

Hans I-Chen Harn et al.

Summary: The study compares laboratory and African spiny mice, revealing a link between Twist1 and differences in hair follicle regenerative behavior after wounding. Through investigation of tissue mechanics and molecular responses, it demonstrates opposing trends in spatial and temporal morphogenetic field formation in these two mouse models following wound stiffness landscapes.
