Crystal structures of human NSDHL and development of its novel inhibitor with the potential to suppress EGFR activity

Dong-Gyun Kim et al.

Summary: NSDHL, a crucial enzyme in human cholesterol synthesis and regulator of EGFR, has been studied for its potential therapeutic applications. Two X-ray crystal structures of human NSDHL were reported, revealing coenzyme-binding details and a conformational change. A novel inhibitor for NSDHL was identified, showing suppressive activity towards EGFR, enhancing antitumor effects in EGFR-driven cancer cells. These findings may serve as a platform for developing therapeutic agents against NSDHL-related diseases.

Posttranslational Regulation of HMG CoA Reductase, the Rate-Limiting Enzyme in Synthesis of Cholesterol

Marc M. Schumacher et al.

Summary: The text discusses the posttranslational regulation mechanisms of HMG-CoA reductase, including sterol-accelerated ubiquitination and ER-associated degradation, augmented by GGpp. By regulating the trafficking of proteins, cells maintain a suitable supply of isoprenoids while avoiding excessive accumulation of cholesterol.

Molecular structures of human ACAT2 disclose mechanism for selective inhibition

Tao Long et al.

Summary: Research on ACAT2 structure reveals similar membrane topology to ACAT1, with a cholesterol entry site and an activation site; Mutations in these entry or activation sites were found to attenuate ACAT2 activity, providing insights for designing new inhibitors for cardiovascular disease prevention.
