Atopic dermatitis: an expanding therapeutic pipeline for a complex disease

Summary: Recent advances in understanding of the complex phenotype and mechanisms underlying atopic dermatitis (AD) have revealed multiple new potential targets for pharmacological intervention. There are now more than 70 new compounds in development, in addition to regulatory approval for several inhibitors in Europe. The potential for a precision medicine approach to enable prevention and more effective long-term control of this complex disease is highlighted.

Summary: ILC2s are shown to synthesize and release acetylcholine during parasitic nematode infection. Exposure to extracts of certain substances or factors in vivo can promote the synthesis of acetylcholine by ILC2s. The ACh derived from ILC2s plays a functional role in expanding ILC2s for maximal induction of type 2 immunity.

NMB-sensitive neurons project to the caudal ventral respiratory group (cVRG) and activation of these neurons leads to sneezing behavior.

A neuro-mesenchymal unit via a brain-adipose circuit controls ILC2s in adipose tissue, affecting metabolism and obesity. This study identifies a complex interaction between neuronal and immune cells in the regulation of host metabolism and obesity.
