Cell type-selective secretome profiling in vivo

Wei Wei et al.

Summary: This study introduced a proximity biotinylation strategy to label, detect, and enrich secreted polypeptides in a cell type-selective manner in mice, resulting in a proteomic atlas of secretomes from various cell types. The research confirmed known cell type-protein pairs, identified new secreted polypeptides distinguishing between cell types, and uncovered a dynamic nutrient-dependent reprogramming of hepatocyte secretome. The strategy enables dynamic and cell type-specific dissection of the plasma proteome and intercellular signaling mediators.

Tunnels for Protein Export from the Endoplasmic Reticulum

I. Raote et al.

Summary: The functions of COPII coat protein complex in eukaryotic protein secretion are conserved, but standard COPII vesicles cannot accommodate the secretion of metazoan-specific cargoes. Metazoans have evolved modules centered on proteins like TANGO1 to create a novel mode of cargo export at the endoplasmic reticulum by interacting with COPII coats and early secretory pathway membranes.

Control of gasdermin D oligomerization and pyroptosis by the Ragulator-Rag-mTORC1 pathway

Charles L. Evavold et al.

Summary: The Ragulator-Rag complex is found to be necessary for GSDMD pore formation and pyroptosis in macrophages, promoting GSDMD oligomerization on the plasma membrane. Defects in GSDMD oligomerization and pore formation can be rescued by mitochondrial poisons that stimulate ROS production, and ROS modulation impacts the ability of inflammasome pathways to promote pore formation downstream of GSDMD cleavage.

Tunneling nanotubes: Reshaping connectivity

Chiara Zurzolo

Summary: Tunneling nanotubes (TNTs) are open membranous channels between cells that allow for the diffusion of various cellular materials. While their discovery has enriched the field of cellular communication research, there is confusion surrounding their structure-function and their ability to transfer material between different cell populations. The use of low-resolution imaging methods in most studies has contributed to this inconsistency, and further research is needed to fully understand the properties of TNTs.

An mTORC1-GRASP55 signaling axis controls unconventional secretion to reshape the extracellular proteome upon stress

Julian Nuechel et al.

Summary: This study identifies mTORC1 as a key regulator of GRASP55 localization in the Golgi through direct phosphorylation, revealing a physiological role for mTORC1 at this organelle. Inhibition of mTORC1 leads to dephosphorylation and relocalization of GRASP55. Multiple proteomic analyses show that many cargoes follow this unconventional secretory route to alter the cellular secretome and surfactome.
