Summary: The embryonic endocardium plays a vital role in early heart development by inducing the formation of trabecular myocardium, the source of valvular cells and part of the coronary vasculature. Human endocardial cells offer unique therapeutic opportunities, such as engineering biological valves and cell-based therapy for damaged hearts. By generating hPSC-derived endothelial cells, key signaling pathways for the development of endocardial cells were identified, with a novel role for BMP10 in lineage specification.

Human heart-forming organoids recapitulate early heart and foregut development

Summary: This study successfully generated three-dimensional heart-forming organoids that closely resemble early native heart anlagen, containing myocardial layer, endocardial-like cells, septum-transversum-like anlagen, and distinct anterior versus posterior foregut endoderm tissues. These organoids were used to study genetic defects in vitro and demonstrated a phenotype similar to cardiac malformations seen in transgenic mice with the knockout of NKX2.5 gene.

Tunable electroconductive decellularized extracellular matrix hydrogels for engineering human cardiac microphysiological systems

Summary: Hybrid hydrogels composed of decellularized porcine myocardial extracellular matrix (dECM) and reduced graphene oxide (rGO) were developed to improve the functionality of cardiomyocytes. These hydrogels can also be used as bioinks to print cardiac tissues in a high-throughput manner. Modulating the composition and properties of the materials can enhance the electrophysiological function of cardiac tissues.

Cardioids reveal self-organizing principles of human cardiogenesis

Summary: Organoids derived from human pluripotent stem cells can spontaneously form chamber-like structures resembling the human heart, which can be controlled and manipulated for studying heart diseases and regeneration. The establishment of self-organizing cardioids provides a powerful platform for mechanistic studies on developmental defects and potential translational research in the future.
