Methanol and methyl group conversion in acetogenic bacteria: biochemistry, physiology and application

Summary: The production of bulk chemicals from petroleum sources leads to greenhouse gas emissions. The development of bio-based processes, particularly using acetogenic bacteria, has gained attention as an alternative. These anaerobic organisms produce acetate and ethanol from CO2, CO, and H2, with methanol also being a suitable substrate. The biochemistry and bioenergetics of methanol conversion in anaerobic bacteria is discussed, along with the possibility of biochemical production from methanol.

Transcriptome and translatome of CO2 fixing acetogens under heterotrophic and autotrophic conditions

Summary: Acetogens utilize gaseous feedstocks to synthesize biomass and metabolites efficiently through the Wood-Ljungdahl pathway, making them a potential platform for biochemical production. Understanding the transcriptional and translational regulation in acetogens during autotrophic growth can aid in designing strains for better productivity. Data on genome-scale transcriptional and translational responses of acetogens during CO2 fixation can help in discovering regulatory elements and engineering strains for desired biochemical production.

Summary: Eubacterium limosum KIST612 primarily produces butyrate from acetate and glucose, with limited production observed from carbon monoxide, hydrogen, CO2, or formate. Interestingly, butyrate formation was also seen when using glucose or methanol as substrates during the stationary growth phase.

Production of the biocommodities butanol and acetone from methanol with fluorescent FAST-tagged proteins using metabolically engineered strains of Eubacterium limosum

Summary: The addition of FAST as an oxygen-independent fluorescent reporter protein expands the genetic toolbox of E. limosum. Our results show that FAST-tagged fusion proteins can be constructed without negatively impacting the stability, functionality, and productivity of the resulting enzyme. Butanol and acetone can be produced from methanol using recombinant E. limosum strains expressing genes encoding fluorescent FAST-tagged fusion proteins.
