pH-Responsive N∧C-Cyclometalated Iridium(III) Complexes: Synthesis, Photophysical Properties, Computational Results, and Bioimaging Application

Anastasia I. Solomatina et al.

Summary: In this study, four iridium complexes containing cyclometallated N^C ligand and various bidentate L^L ligands were synthesized and characterized. These complexes exhibit moderate to strong phosphorescence in the 550-670 nm range, with emission properties strongly dependent on pH. The complexes localize in lysosomes and acidified endosomes within living cells, demonstrating low toxicity.

Photodynamic therapy with mitochondria-targeted biscyclometallated Ir(III) complexes. Multi-action mechanism and strong influence of the cyclometallating ligand

Elisenda Zafon et al.

Summary: Photodynamic therapy is an alternative to traditional chemotherapy that uses photosensitizers activated by light to reduce side effects. Iridium biscyclometallated complexes are promising photosensitizers with exceptional photophysical properties, targeting mitochondria in cancer cells. These complexes have shown moderate cytotoxicity in different human cancer cell lines and exhibit phototoxic activity upon light irradiation.

Monodentately-coordinated bioactive moieties in multimodal half-sandwich organoruthenium anticancer agents

Tasha R. Steel et al.

Summary: Metallodrugs play a central role in the treatment and diagnosis of diseases, and the choice of ligands coordinated to the metal center is crucial for the biological activity of metal-based anticancer agents. Introducing bioactive ligands may lead to synergistic activity between the metal center and the ligand system, and Ru half-sandwich compounds are a promising class of anticancer agents that have been extensively studied.

Bivalent chromatin as a therapeutic target in cancer: An in silico predictive approach for combining epigenetic drugs

Tomas Alarcon et al.

Summary: Tumour cell heterogeneity poses a major obstacle for targeted anti-cancer therapies, with limited strategies available to mitigate its undesirable impact on clinical outcomes. A computational approach has been developed to design combinatorial therapies against chromatin modifiers, with promising results in circumventing therapeutic challenges posed by tumour heterogeneity. Combinatorial strategies involving modulators of histone H3K4 and H3K27 demethylases are predicted to be more effective in managing non-uniform responses of heterogeneous cancer cell populations.

Methodology for Exploring Patterns of Epigenetic Information in Cancer Cells Using Data Mining Technique

Hanan Aljuaid et al.

Summary: Epigenetic changes are crucial in all cancer types, and identifying common patterns across different cancer types could lead to appropriate treatments. A new technique using data mining and clustering methods was proposed to detect common epigenetic patterns in various cancer types, with a success rate of 88% in detecting specific cancer types based on epigenetic patterns. The research demonstrated the existence of common epigenetic patterns across different cancer types, showing potential for targeted treatments.

The language of chromatin modification in human cancers

Shuai Zhao et al.

Summary: The deregulation of chromatin modification underlies numerous oncogenic processes, contributing to cancer development by inducing epigenetic, transcriptomic, and phenotypic alterations. Understanding and developing more effective drugs to target these chromatin-related vulnerabilities will enhance the treatment of human malignancies. This review synthesizes the connections between chromatin modifications and cancer, highlighting options for therapeutic targeting based on recent advances.
