Nonglutaraldehyde crosslinked bioprosthetic heart valves based on 2-iso-cyanatoethyl methacrylate crosslinked porcine pericardium with improved properties of stability, cytocompatibility and anti-calcification

Cheng Zheng et al.

Summary: Valvular heart diseases (VHD) are common in the elderly, with valve replacement being the most effective treatment. Using 2-isocyanatoethyl methacrylate (ICM) as a crosslinking agent can improve the stability of porcine pericardium and potentially be an alternative to glutaraldehyde in bioprosthetic heart valves (BHVs) crosslinking.

Sodium lignosulfonate cross-linked bioprosthetic heart valve materials for enhanced cytocompatibility, improved hemocompatibility, and reduced calcification

Shumang Zhang et al.

Summary: This study aimed to enhance the performance of bioprosthetic heart valves (BHV) by cross-linking porcine pericardium (PP) with sodium lignosulfonate (SLS). The results showed that SLS-crosslinked PP (SLS-PP) had improved cell proliferation, reduced protein adhesion, and decreased calcification compared to glutaraldehyde-crosslinked PP (Glut-PP). SLS is suggested as a potential alternative cross-linker for BHVs.

A method for simultaneously crosslinking and functionalizing extracellular matrix-based biomaterials as bioprosthetic heart valves with enhanced endothelialization and reduced inflammation

Gaoyang Guo et al.

Summary: The study used in-situ polymerization of sulfonic monomers with decellularized extracellular matrix (ECM) for crosslinking and functionalization, providing a potential material for more durable bioprosthetic heart valves. In vitro tests showed promotion of endothelial cell proliferation and migration, while in vivo tests significantly reduced immune response and calcification.

Structural Valve Deterioration Is Linked to Increased Immune Infiltrate and Chemokine Expression

Sabin J. Bozso et al.

Summary: The study indicates that structural valve deterioration of bioprosthetic xenogenic tissue heart valves is associated with increased immune cell infiltration and the co-expression of several chemokines. Following the development of SVD, explanted valves showed significantly increased immune cell infiltration and an elevated chemokine gradient.

Ulvan mediated VE cadherin antibody and REDV peptide co-modification to improve endothelialization potential of bioprosthetic heart valves

Xianbao Liu et al.

Summary: With an aging population and increasing incidence of degenerative valve diseases, the use of bioprosthetic heart valves (BHVs) is on the rise. Current glutaraldehyde cross-linked bioprostheses face durability issues, which can partially be addressed through decellularization and functionalization with ulvan, REDV, and VE-cadherin antibody (U-R-VE) to enhance endothelial cell adhesion and growth while reducing calcification and inflammation. In vitro and in vivo tests demonstrate the effectiveness of this approach in improving the biocompatibility of decellularized porcine pericardium for use in BHVs.
