Quantification of Glomerular Structural Lesions: Associations With Clinical Outcomes and Transcriptomic Profiles in Nephrotic Syndrome

Jeffrey B. Hodgin et al.

Summary: This study used a new digital pathology scoring system to categorize focal segmental glomerulosclerosis (FSGS) and minimal change disease (MCD) patients and identified histologic parameters associated with clinical outcomes and molecular signatures. A glomerular transcriptomic landscape of apolipoprotein L1 in Black patients with focal segmental glomerulosclerosis

Michelle T. McNulty et al.

Summary: APOL1-associated FSGS is the dominant form of FSGS in Black individuals, and studying the transcriptomic landscape of patient kidneys can help uncover genes and molecular behaviors most relevant to the disease. Differences in mitochondrial gene regulation appear to underlie many observed differences between high- and low-risk Black patients with FSGS.

Rho GTPase regulatory proteins in podocytes

Jun Matsuda et al.

Summary: Rho GTPases play a crucial role in regulating the actin cytoskeleton and are implicated in the pathogenesis of proteinuric glomerular diseases. These GTPases are primarily regulated by three families of proteins – GEFs, GAPs, and GDIs – with their upstream regulatory mechanisms largely unknown in podocytes. Further studies are needed to bridge the knowledge gap in the field.

Complexities of the glomerular basement membrane

Richard W. Naylor et al.

Summary: The glomerular basement membrane (GBM) is crucial for kidney filtration, with studies revealing insights into its composition, assembly, developmental transitions, and filtration role. Defects in GBM components are associated with hereditary and acquired diseases, with current treatments focused on reducing intraglomerular pressure and addressing underlying causes. Therapies targeting GBM maintenance and repair could lead to new treatments for GBM-associated diseases.

Sphingosine-1-Phosphate Metabolism and Signaling in Kidney Diseases

Yelena Drexler et al.

Summary: Recent studies have shown that sphingolipids and sphingolipid metabolites play important roles in the pathogenesis and progression of kidney diseases by influencing the intracellular sphingolipid composition of renal cells and the integrity of the glomerular filtration barrier. The sphingolipid metabolite S1P plays a crucial role in renal cells, and aberrant S1P signaling contributes to the pathogenesis and progression of kidney diseases.

Pathophysiology of diabetic kidney disease: impact of SGLT2 inhibitors

Ralph A. DeFronzo et al.

Summary: Diabetic kidney disease is a leading cause of kidney failure globally, with over 50% of individuals entering dialysis or transplant programmes in the USA affected by it. Hyperglycaemia is the primary factor in its development, leading to various pathophysiological disturbances that contribute to progressive glomerular sclerosis and declining renal function. Multiple agents used in combination are likely to be needed to effectively slow the progression of the disease.

Summary: Research has shown that the pyroptosis-related inflammasome pathway plays a crucial role in the development of DKD, and inhibiting its activation can improve the pathological changes associated with the disease. However, current studies on this topic are still relatively superficial and further exploration is needed for a better understanding.

Deficiency of Mitochondrial Glycerol 3-Phosphate Dehydrogenase Exacerbates Podocyte Injury and the Progression of Diabetic Kidney Disease

Hua Qu et al.

Summary: This study highlights the importance of mitochondrial glycerol 3-phosphate dehydrogenase (mGPDH) in regulating podocyte function and glomerular disease. Inhibition of RAGE or its ligand, S100A10, protects against impaired mitochondrial bioenergetics caused by mGPDH knockdown. Deletion of RAGE in podocytes attenuates nephropathy progression in mGPDH-deficient diabetic mice. Rescuing podocyte mGPDH expression in mice with established glomerular injury significantly improves renal function.
