Dual enzyme-mimic nanozyme based on single-atom construction strategy for photothermal-augmented nanocatalytic therapy in the second near-infrared biowindow

Summary: Nanozyme-based catalytic therapy plays an important role in tumor therapy by generating lethal reactive oxygen species. However, most nanozymes have mono catalytic performances in the tumor microenvironment, which lowers their therapeutic efficiency. In this study, a newly-developed nanozyme with dual enzyme-mimic catalytic activity is designed, which can achieve an enhanced nanocatalytic therapy effect through photothermal augmentation.

Summary: FePc/HNCSs possess peroxidase (POD) and catalase (CAT)-like activities, converting endogenous hydrogen peroxide (H2O2) into highly toxic hydroxyl radicals (OH) for catalytic therapy and decomposing H2O2 into oxygen (O-2) to enhance O-2-dependent PDT. Their enzyme-like activities are significantly enhanced under light irradiation, achieving a high tumor inhibition rate of 96.3% by combining with excellent photothermal effect.

It can provide oxygen, consume glucose, and convert light into heat, showing promising effects in primary tumor and metastatic tumor treatment.

Summary: Two donor-acceptor-donor (D-A-D) structured NIR dyes, BBTT and SeBTT, were designed, with SeBTT NPs showing stronger NIR absorbance and higher photothermal conversion efficiency. In vivo experiments demonstrated that SeBTT NPs can be used as a high contrast photoacoustic imaging agent and effectively suppress tumors under NIR photoirradiation.
