Integrated treatment of perennial ryegrass: Structural characterization of hemicelluloses and improvement of enzymatic hydrolysis of cellulose

Shao-Fei Sun et al.

Summary: By integrating ultrasonic and hydrothermal pretreatments with sequential alkali post extractions, hemicelluloses were successfully isolated and characterized from perennial ryegrass, leading to an improvement in the enzymatic hydrolysis efficiency of cellulose. The collaborative utilization of hemicelluloses and cellulose in ryegrass resulted in the effective removal of amorphous hemicelluloses and lignin, significantly increasing the cellulose enzymatic hydrolysis rate from 43.8 to 91.1%.

Effect of integrated treatment on improving the enzymatic digestibility of poplar and the structural features of isolated hemicelluloses

Shao-Chao Sun et al.

Summary: In this study, a two-step hydrothermal pretreatment combined with alkali extraction method was used to deconstruct poplar cell walls for enzymatic hydrolysis, resulting in the removal of a significant amount of hemicelluloses and lignin. Confocal Raman microscopy revealed that hemicelluloses were unevenly removed from cell walls, mainly originating from the secondary wall. Water-soluble hemicelluloses were found to have a more branched structure compared to alkali-soluble hemicelluloses, with a linear backbone of (1 -> 4)-beta-D-Xylp and 4-O-Me-alpha-d-GlcpA side chains identified in hemicelluloses isolated from poplar. This work provides an efficient pathway to convert poplar into fermentable sugars and hemicelluloses with significant yield.

Recent advances in lignocellulose prior-fractionation for biomaterials, biochemicals, and bioenergy

Xiaojun Shen et al.

Summary: Lignocellulosic biomass is considered the best candidate for producing biomaterials, biochemicals, and bioenergy due to over-consumption of fossil resources and environmental problems. Efficient and green fractionation of lignocellulose components can greatly enhance high-value utilization and biorefinery development, but the heterogeneity of lignocellulosic structure poses challenges to this process. Through strategies such as cellulose-first, hemicelluloses-first, and lignin-first fractionation, selective dissociation and transformation of components in lignocellulose can be achieved, offering both challenges and opportunities for biorefining valorization.

Revealing the topochemical and structural changes of poplar lignin during a two-step hydrothermal pretreatment combined with alkali extraction

Shao-Chao Sun et al.

Summary: This study successfully extracted lignin with high yield and ultra-high purity from poplar using a combined method of hydrothermal pretreatment and alkali extraction. Confocal Raman microscopy analysis revealed that lignin was preferentially delignified in the secondary wall and compound middle lamella, then throughout the entire morphological regions of the cell wall. NMR results showed that the cleavage of beta-O-4 linkages was the dominant reaction, leading to an increase in phenolic hydroxyl groups, a decrease in molecular weight, and heterogeneity of the isolated lignin.
