JASPAR 2022: the 9th release of the open-access database of transcription factor binding profiles

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

EphA2 super-enhancer promotes tumor progression by recruiting FOSL2 and TCF7L2 to activate the target gene EphA2

Summary: This study identified a new super-enhancer EphA2-SE in various tumor cells, which drives the expression of oncogene EphA2 by recruiting FOSL2 and TCF7L2 transcription factors. Deletion of EphA2-SE inhibited tumor growth and metastasis by blocking PI3K/AKT and Wnt/β-catenin pathway, and overexpression of EphA2 rescued the effect of deletion. Animal models further confirmed that EphA2-SE deletion suppressed tumor proliferation in vivo.
