Membrane Permeability in Cyclic Peptides is Modulated by Core Conformations

Flaviu Cipcigan et al.

Summary: The study using cyclic pentapeptides found that traditional physicochemical criteria of permeable peptides were violated, suggesting counter-intuitive strategies may be needed to enhance permeability. Furthermore, core globularity was identified as a promising descriptor that could expand the capability of standard predictive models.

Geometrically Diverse Lariat Peptide Scaffolds Reveal an Untapped Chemical Space of High Membrane Permeability

Colin N. Kelly et al.

Summary: This study focused on investigating the structure-permeability relationships in lariat peptides, which are cyclized from one termini onto a side chain. A library of over 1000 novel lariat peptide scaffolds with molecular weights around 1000 was generated using stable isotopes to encode stereochemistry. The research found that passive permeability of lariat peptides was strongly dependent on N-methylation, stereochemistry, and ring topology, revealing various trends in structure-permeability relationships.

Connecting Hydrophobic Surfaces in Cyclic Peptides Increases Membrane Permeability

Huy N. Hoang et al.

Summary: Research has shown that a single methyl group only increases membrane permeability when it connects or expands hydrophobic surface patches. Positional isomers with the same characteristics can have different permeabilities based on the size of the largest continuous hydrophobic surface patch, illuminating a key local molecular determinant of membrane permeability.

Cyclosporin Structure and Permeability: From A to Z and Beyond

Karen M. Corbett et al.

Summary: Cyclosporins are a class of peptides with wide pharmaceutical applications, some of which exhibit high membrane permeability not easily explained by structural metrics. Experimental studies have been conducted to understand the structure and permeability of cyclosporins, including their interaction with metal complexes. Models have been proposed to explain the conformation-dependent permeability of cyclosporins and similar compounds, with gaps in current knowledge identified for further exploration.

Macrocyclic Peptides as a Novel Class of NNMT Inhibitors: A SAR Study Aimed at Inhibitory Activity in the Cell

Kyohei Hayashi et al.

Summary: Utilizing in vitro translated macrocyclic peptide libraries, a novel class of NNMT inhibitors was identified, with dramatic improvement in inhibitory activity based on X-ray cocrystal structures. Through balancing lipophilicity and biological activity, inhibitory activity against NNMT in cell-based assay was successfully achieved. These findings highlight the potential of cyclic peptides as a new drug discovery modality even for intracellular targets.
