Microglia Elimination Increases Neural Circuit Connectivity and Activity in Adult Mouse Cortex

Yong-Jun Liu et al.

Summary: Microglia play a crucial role in sculpting neuronal circuit connectivity and regulating subsequent functional activity in the adult cortex. Depletion of microglia increases synaptic connections and enhances neural activities, which recover following re-population of adult microglia.

The Multifaceted Roles of KCC2 in Cortical Development

Mari A. Virtanen et al.

Summary: KCC2 is a multifunctional molecule that can regulate the strength and polarity of GABAergic currents and cytoskeletal dynamics. Its various splice variants play roles in controlling early network events and the formation and plasticity of cortical dendritic spines. The versatility of KCC2 actions is evident in mature neurons during plasticity, disease, and aging, making it one of the most important molecules shaping the overall neuronal phenotype.

GABA-receptive microglia selectively sculpt developing inhibitory circuits

Emilia Favuzzi et al.

Summary: Microglia, the resident immune cells of the brain, selectively interact with inhibitory cortical synapses during a critical window of mouse postnatal development through initiating a transcriptional synapse remodeling program with GABA, demonstrating the importance of selective communication between neuronal and glial cell types in brain wiring.

Electrophysiology of ionotropic GABA receptors

Erwan Sallard et al.

Summary: This review discusses the major and diverse roles of GABA(A) receptors in regulating neuronal communication and brain functioning, emphasizing their complex electrophysiological properties and diverse subtypes. The diversity of GABA(A) receptors widens the neuronal responses to external signals and contributes to shaping the electrical activity of neurons and other cell types.

A subthreshold synaptic mechanism regulating BDNF expression and resting synaptic strength

Patricia M. Horvath et al.

Summary: Recent studies have shown that protein translation can be regulated by spontaneous excitatory neurotransmission, while the impact of spontaneous neurotransmitter release on gene transcription remains unclear. Blocking spontaneous inhibitory events increases the transcription of BDNF and Npas4 through altered synaptic calcium signaling, which can be prevented by antagonism of NMDA receptors or L-type voltage-gated calcium channels. Transcription can be bidirectionally altered by manipulating spontaneous inhibitory, but not excitatory, currents. Moreover, blocking spontaneous inhibitory events results in multiplicative downscaling of excitatory synaptic strength, dependent on transcription and BDNF signaling. These findings highlight the role of spontaneous inhibitory neurotransmission in setting excitatory synaptic strength at rest.

Adhesion dynamics in the neocortex determine the start of migration and the post-migratory orientation of neurons

Ekaterina Epifanova et al.

Summary: Neuronal adhesion is regulated by Zeb2 to control the initiation and orientation of radial migration in the neocortex, involving pathways regulated by Neuropilin-1 and Caadherin-6. Neurons undergo an independent orientation process after migration is completed.

Neuromodulation by the immune system: a focus on cytokines

Andrea Francesca Salvador et al.

Summary: Interactions between the immune system and the nervous system play a crucial role in shaping host behavior and nervous system function. These interactions may have evolved to maximize an organism's ability to respond to environmental threats. The immune system relays information to the nervous system through cytokine signaling, and the brain may be hardwired to process information from the immune system in a specific way.
