Mitochondrial dysfunctions in neurodegenerative diseases: role in disease pathogenesis, strategies for analysis and therapeutic prospects

Federica Rey et al.

Summary: This review discusses the role of mitochondrial impairment in the development of neurodegenerative diseases, focusing on Amyotrophic Lateral Sclerosis, Parkinson's disease, and Alzheimer's disease. It highlights the importance of mitochondrial fusion, fission, reactive oxygen species production, and energy failure in these diseases. The review also explores methods to measure mitochondrial health and potential therapeutic strategies to mitigate mitochondrial dysfunction.

Mitochondrial Dysfunction and Mitophagy in Parkinson's Disease: From Mechanism to Therapy

Ana Belen Malpartida et al.

Summary: Mitochondrial dysfunction has long been associated with Parkinson's disease, although its exact role remains unclear. Recent discoveries on the role of PD familial genes in mediating mitophagy and the overlap with sporadic PD have opened up new opportunities for therapeutically targeting mitochondria in PD. Progress has also been made in understanding the upstream and downstream regulators of canonical and noncanonical PINK1/parkin-mediated mitophagy in response to mitochondrial damage.

14-3-3 mitigates alpha-synuclein aggregation and toxicity in the in vivo preformed fibril model

Rachel Underwood et al.

Summary: Alpha-synuclein is a key component in synucleinopathies like Parkinson's Disease and Dementia with Lewy Bodies, and its transmission can be influenced by the neuroprotective role of 14-3-3 theta in reducing aggregation and intercellular spread of alpha syn.

The importance of ultrastructural analysis of memory

Malgorzata Borczyk et al.

Summary: This study compares the effects of long-term potentiation of synaptic transmission (LTP) and behavioral training on glutamatergic synapses in the hippocampus at the ultrastructural level, using electron microscopy. It is found that while LTP induces growth of dendritic spines and post-synaptic densities (PSD), behavioral training results in transient and long-lasting changes in synaptogenesis and PSD volume, indicating potential long-term enhancement of synaptic functions. Additionally, formation of multi-innervated spines (MIS) is linked to long-term memory, suggesting the importance of ultrastructural analysis in revealing synaptic changes associated with long-term information storage in the brain.
