Abnormalities of synaptic mitochondria in autism spectrum disorder and related neurodevelopmental disorders

Liliana Rojas-Charry et al.

Summary: Autism spectrum disorder (ASD) is a neurodevelopmental condition characterized by impairments in social interaction and repetitive behaviors, with mitochondrial dysfunction considered as an important underlying pathogenic factor. However, the pathophysiology of ASD is still not fully understood, with mutations in synaptic genes also playing a role in the disorder. Mitochondria play diverse functions at the synapse, beyond their classical role as energy providers.

Acetyl-CoA flux from the cytosol to the ER regulates engagement and quality of the secretory pathway

Inca A. Dieterich et al.

Summary: N epsilon-lysine acetylation in the ER is crucial for quality control, with dysfunction of AT-1 leading to severe disease phenotypes. Two models of AT-1 dysregulation showed reorganization of the ER, ERGIC, and Golgi apparatus, highlighting the importance of AT-1 in maintaining proper organization and engagement of the secretory pathway.

Emerging behavioral and neuroimaging biomarkers for early and accurate characterization of autism spectrum disorders: a systematic review

Chandrakanta S. Hiremath et al.

Summary: Early identification and characterization of ASD features can lead to early treatment and better outcomes. Various tools and methods have been developed for this purpose, with research focusing on biomarkers in ASD. Advanced assessment tools like MRI and behavioral methods are being used to accurately characterize ASD features and facilitate targeted therapy programs. Longitudinal research studies are needed for accurate characterization of brain-behavioral changes over time.

ATG9A regulates proteostasis through reticulophagy receptors FAM134B and SEC62 and folding chaperones CALR and HSPB1

Brendan K. Sheehan et al.

Summary: The acetylation of ATG9A within the ER lumen is regulated by AT-1/SLC33A1, and dysregulation of AT-1 activity can lead to severe disease phenotypes. The induction of reticulophagy requires interactions between ATG9A, FAM134B, and SEC62 on the cytosolic side of the ER. Furthermore, ATG9A associates with various proteins that help maintain ER proteostasis.

Endoplasmic reticulum acetyltransferases Atase1 and Atase2 differentially regulate reticulophagy, macroautophagy and cellular acetyl-CoA metabolism

Michael J. Rigby et al.

Summary: Rigby et al. utilize Atase1 or Atase2-deficient transgenic mice to demonstrate the distinct roles of these endoplasmic reticulum acetyltransferases in regulating reticulophagy and macroautophagy. Their findings highlight Atase1 as a potential translational target for diseases characterized by toxic protein aggregation in the secretory pathway.
