Antagonism Between PEDF and TGF-β Contributes to Type VI Osteogenesis Imperfecta Bone and Vascular Pathogenesis

This study investigates the role of pigment epithelium-derived factor (PEDF) in OI, specifically in the severe type VI form. The findings demonstrate that loss of PEDF delays osteoblast maturation and extracellular matrix mineralization, leading to the accumulation of unmineralized osteoid and abnormal bone lamellae. Additionally, PEDF deficiency results in increased bone vascularization through the activation of angiogenesis and the involvement of specific endothelial cells. The study also reveals a functional antagonism between PEDF and TGF-beta signaling pathways, highlighting their role in controlling osteogenesis and bone vascularization in type VI OI. These findings contribute to a better understanding of the disease mechanism and offer potential therapeutic targets for type VI OI.
