A Simple Heat Treatment Increases SpCas9-Mediated Mutation Efficiency in Arabidopsis

Shuta Kurokawa et al.

Summary: The CRISPR/Cas9 system is commonly used for genome editing in plants like Arabidopsis, rice, and tobacco. Efficiency of mutation in Arabidopsis using SpCas9 can be improved by using the parsley UBIQITIN promoter and a simple heat treatment at 37 degrees C for 24 hours. This method showed a significant increase in mutation efficiency without off-target mutations.
