Early truncation of the N-terminal variable region of EYA4 gene causes dominant hearing loss without cardiac phenotype

Summary: This study reports a Chinese family with autosomal dominant progressive hearing loss carrying a novel nonsense mutation of the EYA4 gene. Despite the mutation, affected members did not show any cardiac abnormalities during screening. The overall expression level of the EYA4 gene was found to be lower in the proband compared to unaffected relatives, highlighting the need for further research to understand the complex genotype-phenotype correlation of EYA4 mutations.

Genome-first approach to rare EYA4 variants and cardio-auditory phenotypes in adults

Summary: The study found that EYA4 gene burden is significantly associated with audiometric proven HL, Mobitz Type II AV block, and syndromic presentation of HL and primary cardiomyopathy. These associations were more subtle cardio-auditory phenotypes in an adult medical biobank population, which have not been previously reported. The findings suggest a role for genetic testing in adults presenting with hearing loss.
