RIP3 blockade prevents immune-mediated hepatitis through a myeloid-derived suppressor cell dependent mechanism

Man Liu et al.

Summary: The study found that increased expression and activation of RIP3 in livers of autoimmune hepatitis patients and animal models, and inhibition of RIP3 kinase prevented immune-mediated hepatitis by promoting MDSCs infiltration with immunoregulatory properties. This indicates that RIP3 blockade may be a novel avenue for autoimmune hepatitis treatment.

Effect of Coptis chinensis franch and Magnolia officinalis on intestinal flora and intestinal barrier in a TNBS-induced ulcerative colitis rats model

Qian Xie et al.

Summary: In this study, the therapeutic effects of Coptis chinensis Franch and Magnolia officinalis on TNBS-induced ulcerative colitis in rats were investigated. It was found that these herbal medicines could improve disease indicators, reduce colonic mucosal damage, and regulate intestinal flora. They also showed anti-inflammatory effects and protected the integrity of the tight junction, indicating their potential in preventing intestinal barrier damage.

ABIN3 Negatively Regulates Necroptosis-induced Intestinal Inflammation Through Recruiting A20 and Restricting the Ubiquitination of RIPK3 in Inflammatory Bowel Disease

Mingxia Zhou et al.

Summary: This study found that excessive necroptosis plays an important role in uncontrolled inflammation and intestinal barrier defects in IBD and experimental colitis. ABIN3 regulates inflammatory response and intestinal barrier function by interacting with A20 and coordinating the K63 deubiquitination modification of necroptosis.

Inhibition of neuronal necroptosis mediated by RIP1/RIP3/MLKL provides neuroprotective effects on kaolin-induced hydrocephalus in mice

Chang Liu et al.

Summary: In a hydrocephalus mouse model, the necroptosis pathway inhibitor Nec-1 and GSK872 showed neuroprotective effects by reducing necrotic cell death, attenuating levels of p-RIP3 and p-MLKL, slightly improving neurobehaviors, and inhibiting microglial and astrocyte activation and inflammation. RIP1/RIP3/MLKL pathway mediates necroptosis in the cortex and hippocampus.
