Cancer Statistics, 2021

Rebecca L. Siegel et al.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Short-course radiotherapy followed by chemotherapy before total mesorectal excision (TME) versus preoperative chemoradiotherapy, TME, and optional adjuvant chemotherapy in locally advanced rectal cancer (RAPIDO): a randomised, open-label, phase 3 trial

Renu R. Bahadoer et al.

Summary: The RAPIDO trial aimed to reduce systemic relapses in locally advanced rectal cancer through short-course radiotherapy, chemotherapy, and delayed surgery. Results showed a decreased probability of disease-related treatment failure in the experimental group compared to standard care, with diarrhea being the most common adverse event and neurological toxicity being prominent in the standard care group during adjuvant chemotherapy.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Neoadjuvant chemotherapy with FOLFIRINOX and preoperative chemoradiotherapy for patients with locally advanced rectal cancer (UNICANCER-PRODIGE 23) : a multicentre, randomised, open-label, phase 3 trial

Thierry Conroy et al.

Summary: In patients with cT3 or cT4 M0 rectal cancer, intensifying chemotherapy with FOLFIRINOX before preoperative chemoradiotherapy significantly improved disease-free survival outcomes and reduced neurotoxicity. Thus, the PRODIGE 23 results have the potential to impact clinical practice.

Detection of low-frequency DNA variants by targeted sequencing of the Watson and Crick strands

Joshua D. Cohen et al.

Summary: The SaferSeqS method efficiently detects low-frequency mutations by introducing identical molecular barcodes and enriching target sequences with strand-specific PCR, achieving high sensitivity and specificity, and detecting variants at frequencies below 1 in 100,000 DNA template molecules with a background mutation rate of <5 x 10(-7) mutants per base pair (bp).

Conventional myelosuppressive chemotherapy for non-haematological malignancy disrupts the intestinal microbiome

Lito E. Papanicolas et al.

Summary: Conventional myelosuppressive chemotherapy results in significant changes in gut microbiota characteristics during the predicted myelosuppression period post-chemotherapy, but these changes are no longer significant at the end of the chemotherapy cycle. Further research is needed to link microbiome changes during chemotherapy to clinical outcomes.

International consensus recommendations on key outcome measures for organ preservation after (chemo)radiotherapy in patients with rectal cancer

Emmanouil Fokas et al.

Summary: The increasing consideration of organ preservation in multimodal treatment strategies for rectal cancer aims to reduce surgical morbidities and maintain patients' anorectal function and quality of life. However, standardization of key outcome measures is currently lacking, requiring further research and consensus recommendations.
