Multiple Topoisomerase I (TopoI), Topoisomerase II (TopoII) and Tyrosyl-DNA Phosphodiesterase (TDP) inhibitors in the development of anticancer drugs

Emma Baglini et al.

Summary: DNA topoisomerases are crucial enzymes involved in DNA topology regulation, with inhibitors targeting TopoI and TopoII showing broad-spectrum antitumor activities and potential to reduce drug resistance and side effects in cancer treatment. Targeting both enzymes simultaneously could provide more efficient and safe drugs for clinical usage.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Synthesis of novel potent cytotoxicy podophyllotoxin-naphthoquinone compounds via microwave-assited multicomponent domino reactions

Quynh Giang Nguyen Thi et al.

Summary: A series of novel podophyllotoxin-naphthoquinone compounds were synthesized in good yields and evaluated for cytotoxicity activities. Some of the compounds showed high inhibitory activities against cancer cell lines with low toxicity for non-cancerous cell lines, indicating their potential importance in the development of anticancer agents.

The ester derivatives obtained by C-ring modification of podophyllotoxin induce apoptosis and inhibited proliferation in PC-3M cells via down-regulation of PI3K/Akt signaling pathway

Yan Zhao et al.

Summary: In this study, 20 ester derivatives of Podophyllotoxin (PPT) were designed and synthesized, with V-05 showing the strongest inhibitory effect on PC-3M cells by regulating apoptotic signaling pathways. This research provides insights for the potential application of PPT derivatives in effective medicine development for human prostate cancer treatment.

Metabolic map of the antiviral drug podophyllotoxin provides insights into hepatotoxicity

Dongxue Sun et al.

Summary: Podophyllotoxin (POD) is a natural compound with antiviral and anticancer activities. This study identified a total of 38 metabolites of POD in vitro and in vivo, with metabolism mainly dependent on enzymes such as CYP2C9, CYP2C19, CYP3A4, and CYP3A5. The study also found that POD, and not its metabolites, induced hepatic and intestinal toxicities in mice.
