Retinoid X Receptor: Cellular and Biochemical Roles of Nuclear Receptor with a Focus on Neuropathological Involvement

Samridhi Sharma et al.

Summary: Retinoid X receptors (RXRs) play critical roles in various neurological disorders by regulating gene expression and influencing neurodevelopment, metabolism, and biochemical processes. Abnormal RXR signaling affects neuronal stress and neuroinflammatory networks. Targeting RXRs through pharmacological ligands has shown protective effects in neuronal injury models. This review summarizes the roles of RXR, its interacting partners, and ligands in CNS disorders, providing insights for further research.

TBC1D3 promotes neural progenitor proliferation by suppressing the histone methyltransferase G9a

Qiong-Qiong Hou et al.

Summary: The study discovered an interaction between TBC1D3 and G9a, where TBC1D3 inhibits the histone methyltransferase activity of G9a to promote neural progenitor proliferation, leading to cortical expansion. The findings demonstrate a mechanism by which a hominoid-specific gene contributes to the growth of the cerebral cortex.

Single-cell epigenomics reveals mechanisms of human cortical development

Ryan S. Ziffra et al.

Summary: During mammalian development, differences in chromatin state play a crucial role in cell fate specification and defining cell identity in the developing brain. Through studying chromatin accessibility patterns, it was found that cerebral organoids recapitulate most cell-type-specific enhancer accessibility patterns but lack some open chromatin regions found in vivo. Systematic comparison of chromatin accessibility across brain regions revealed unexpected diversity among neural progenitor cells and highlighted the role of retinoic acid signaling in neuronal lineage specification in the prefrontal cortex.

Genome-wide enhancer maps link risk variants to disease genes

Joseph Nasser et al.

Summary: The study utilized the ABC model to create enhancer-gene maps in 131 human cell types and tissues and interpreted the functions of GWAS variants in 72 diseases and complex traits. It was found that in inflammatory bowel disease, causal variants are enriched in predicted enhancers in specific cell types, and the ABC model shows higher precision in connecting noncoding variants to target genes.

Detection of Alu Exonization Events in Human Frontal Cortex From RNA-Seq Data

Liliana Florea et al.

Summary: Alu exonization events contribute to 5% of alternatively spliced exons in the human genome, and computational methods have been developed to detect Alu sequences in mRNA transcripts. Analysis of 117 GTEx human frontal cortex samples revealed hundreds of Alu-containing mRNAs, including both fixed Alu loci and polymorphic Alu elements. This unique resource provides valuable insights into the characteristics and tissue-specific expression patterns of Alu-containing mRNAs.
