Cryo-EM structure of the full-length WzmWzt ABC transporter required for lipid-linked O antigen transport

Christopher A. Caffalette et al.

Summary: O antigens play a crucial role in gram-negative bacteria by forming serotype-specific cell surface barriers. The WzmWzt ABC transporter facilitates the assembly, modification, and translocation of O antigens to the cell surface in a process that modulates ATPase activity. The cryo-electron microscopy structure of WzmWzt provides insights into the asymmetric organization of the transporter and its closed conformation that likely regulates O antigen recruitment and translocation.

Multidrug Resistance in Mammals and Fungi-From MDR to PDR: A Rocky Road from Atomic Structures to Transport Mechanisms

Narakorn Khunweeraphong et al.

Summary: Multidrug resistance can be a major issue in cancer treatment and microbial infections, with dysregulated overexpression of ATP-binding cassette transporter members being linked to this phenomenon. High-resolution atomic structures from X-ray crystallography or cryo-EM are helping to improve understanding of the mechanisms and dynamics driving these transporters, offering new perspectives for therapeutic interventions.

Structural Basis of Drug Recognition by the Multidrug Transporter ABCG2

Julia Kowal et al.

Summary: In this study, three cryo-EM structures of human ABCG2 bound to anticancer drugs tariquidar, topotecan, and mitoxantrone were presented, revealing the binding pocket details and functional importance of key residues. Mutagenesis experiments and 3D variability analysis provided further insights into the mechanisms by which ABCG2 distinguishes between inhibitors and substrates.
