The β-Secretase BACE1 in Alzheimer's Disease

Harald Hampel et al.

Summary: BACE1 is a well-validated therapeutic target for slowing down Aβ production in early AD, despite previous clinical trials being discontinued. Further research should focus on the roles of BACE1 and Aβ in physiological and pathophysiological processes, as well as the substrates and downstream effects of BACE1 and BACE2.

Lessons Learnt from the Second Generation of Anti-Amyloid Monoclonal Antibodies Clinical Trials

Angela Tian Hui Kwan et al.

Summary: This paper reviews recent anti-A beta passive monotherapies that have progressed to phase 3 clinical trials, discussing four clinical trial programs targeting different forms of A beta using exogenous monoclonal antibodies. The paper concludes with potential reasons for their lack of success and the lessons learned from these trials. Future disease-modifying trials for AD should focus on asymptomatic, A beta-positive individuals and explore potential benefits of anti-A beta and anti-tau drug combinations.

Findings of Efficacy, Safety, and Biomarker Outcomes of Atabecestat in Preclinical Alzheimer Disease A Truncated Randomized Phase 2b/3 Clinical Trial

Reisa Sperling et al.

Summary: This study reveals that atabecestat treatment in preclinical Alzheimer disease patients is associated with dose-related cognitive worsening and neuropsychiatric adverse events. However, after stopping treatment, cognitive status returned to baseline and the frequency of adverse events returned to placebo levels.

Aminopeptidase A contributes to biochemical, anatomical and cognitive defects in Alzheimer's disease (AD) mouse model and is increased at early stage in sporadic AD brain

Audrey Valverde et al.

Summary: Research has shown that aminopeptidase A plays a key role in the N-terminal truncation of A beta and is involved in Alzheimer's disease related anatomical and behavioral defects. Targeting this enzyme may be beneficial in combating AD pathology.

The crystal structure of a 250-kDa heterotetrameric particle explains inhibition of sheddase meprin ß by endogenous fetuin-B

Ulrich Eckhard et al.

Summary: The interaction between M ss and FB is analyzed in this study, revealing how FB stabilizes the enzyme and inhibits its activity with high affinity. The crystal structure shows that FB blocks the catalytic activity of M ss through a unique structural mechanism, preventing cleavage of substrates.

Phosphorylation of meprin β controls its cell surface abundance and subsequently diminishes ectodomain shedding

Fred Armbrust et al.

Summary: Meprin beta is a zinc-dependent metalloprotease with a unique cleavage specificity for acidic amino acids at the cleavage site. Phosphorylation of meprin beta affects its cell surface activity and substrate cleavage extent, leading to internalization and degradation. The involvement of PKC isoforms in meprin beta phosphorylation was also demonstrated in this study.
