In Vitro Biosynthetic Pathway Investigations of Neuroprotectin D1 (NPD1) and Protectin DX (PDX) by Human 12-Lipoxygenase, 15-Lipoxygenase-1, and 15-Lipoxygenase-2

Wan-Chen Tsai et al.

Summary: This study investigated the reactions of human platelet 12-lipoxygenase, human reticulocyte 15-lipoxygenase-1, and human epithelial 15-lipoxygenase-2 with docosahexaenoic acid to produce 17S-HpDHA. The enzymes showed varying efficiencies and product distributions, with 15-LOX isozymes producing higher levels of 17S-HpDHA. The differences in the effective k(cat)/K-M values for 17S-HpDHA suggest distinct reactivity patterns among the enzymes.

Molecular insights into lipoxygenases for biocatalytic synthesis of diverse lipid mediators

Jung-Ung An et al.

Summary: Oxylipins derived from C20- and C22-polyunsaturated fatty acids, known as lipid mediators (LMs), are crucial signaling molecules involved in maintaining homeostasis and promoting healing responses to infection and inflammation in humans. Specialized pro-resolving mediators (SPMs) are a subset of LMs that play a key role in resolving inflammation and infection by activating phagocytosis. Microbial LOXs offer the potential for more efficient synthesis of human LMs from PUFAs, with increased activity and stability compared to human LOXs.
