Doxorubicin-Resistant TNBC Cells Exhibit Rapid Growth with Cancer Stem Cell-like Properties and EMT Phenotype, Which Can Be Transferred to Parental Cells through Autocrine Signaling

Anjugam Paramanantham et al.

Summary: Research suggests that breast cancer stem cells and epithelial-mesenchymal transition may play a role in resistance to doxorubicin. By characterizing doxorubicin-resistant MDA-MB-231 cells, it was found that they exhibit EMT features and CSC properties, which can be transferred to parental cells through autocrine signaling. The up-regulation of EGFR and its downstream signaling, as well as increased AKT and ERK1/2 expression, were associated with the doxorubicin resistance of DRM cells.

Assessment of azithromycin as an anticancer agent for treatment of imatinib sensitive and resistant CML cells

Tulin Ozkan et al.

Summary: The study suggests that single AZT treatment or combined AZT/IMA therapy can inhibit cell proliferation, induce apoptosis, and increase caspase 3/7 activity in both K562S and high MDR-1 (Pgp) expressing K562R cells. Additionally, the combination of AZT and IMA can decrease Pgp function in K562R cells and significantly increase the BAX/BCL-2 ratio in both K562S and K562R cells.

Mitochondrial ATP fuels ABC transporter-mediated drug efflux in cancer chemoresistance

Emily L. Giddings et al.

Summary: Chemotherapy is the standard treatment for most cancers worldwide, but chemoresistance due to ABC transporters is a significant challenge. The study highlights the role of mitochondrial respiration in supplying ATP for ABC transporters, and how inhibiting mitochondrial respiration can overcome chemoresistance. Development of MCJ mimetics offers a potential alternative strategy for treating multiple cancers by targeting this mechanism.

Inhibition of mitochondrial translation suppresses glioblastoma stem cell growth

Denise Sighel et al.

Summary: This study suggests that targeting mitochondrial translation could be a potential therapeutic strategy to suppress GSC growth in GBM. The bacterial antibiotic quinupristin/dalfopristin (Q/D) was found to effectively inhibit GSC growth by binding to the large mitoribosomal subunit, inhibiting mitochondrial protein synthesis, and disrupting OXPHOS complexes.

Binding and Action of Triphenylphosphonium Analog of Chloramphenicol upon the Bacterial Ribosome

Chih-Wei Chen et al.

Summary: The semi-synthetic triphenylphosphonium analog of chloramphenicol, CAM-C4-TPP, shows stronger affinity and higher inhibitory potency towards bacterial ribosomes than chloramphenicol. Its X-ray crystal structure reveals the compound's unique binding mode and potential as a chemical scaffold for further development in medicinal chemistry.

Triphenilphosphonium Analogs of Chloramphenicol as Dual-Acting Antimicrobial and Antiproliferating Agents

Julia A. Pavlova et al.

Summary: This study synthesized and studied new chimeric compounds that combine chloramphenicol and triphenylphosphonium, showing dual action targeting both bacterial ribosomes and cellular membranes. These compounds demonstrated inhibitory effects on CHL-resistant bacterial strains in addition to suppressing protein synthesis.
