Ferroptosis and its potential role in the physiopathology of Parkinson's Disease

Laura Mahoney-Sanchez et al.

Summary: Parkinson's Disease is a common neurodegenerative disorder with motor impairments and non-motor symptoms. Pathological features include dopaminergic neuronal death, alpha-synuclein deposition, iron accumulation, oxidative stress, and lipid peroxidation damage. Targeting pathways like ferroptosis could have therapeutic potential in treating PD.

Summary: The overexpression of SLC7A11 promotes tumor growth while causing significant metabolic reprogramming costs for cancer cells. Cancer cells with high expression of SLC7A11 exhibit glucose- and glutamine-dependency, presenting potential metabolic vulnerabilities for therapeutic targeting.

Resolving the paradox of ferroptotic cell death: Ferrostatin-1 binds to 15LOX/PEBP1 complex, suppresses generation of peroxidized ETE-PE, and protects against ferroptosis

Tamil S. Anthonymuthu et al.

Summary: HpETE-PE is a cell death signal in ferroptosis, produced by the 15LOX/PEBP1 complex. Fer-1 does not directly affect 15LOX, but can effectively inhibit the production of HpETE-PE by the 15LOX/PEBP1 complex. This study resolves the long-standing Fer-1 anti-ferroptotic paradox.

Cellular degradation systems in ferroptosis

Xin Chen et al.

Summary: Ferroptosis is an iron-dependent oxidative cell death driven by lipid peroxidation. Various antioxidant systems play a significant role in preventing ferroptotic death. The cellular degradation systems play a dual role in regulating the ferroptotic response.

The Multifaceted Regulation of Mitochondria in Ferroptosis

Hao Wu et al.

Summary: Ferroptosis is a novel form of regulated cell death initiated by lethal lipid peroxidation catalyzed by cellular labile free iron. It differs significantly from other regulated cell death forms at morphological, genetic, and biochemical levels. Dysregulated ferroptosis is highly relevant to the pathogenesis of degenerative diseases and organ injuries in human patients, and the targeted induction of ferroptosis is considered a potential therapeutic strategy for therapy-resistant cancers. Mitochondria play a crucial role in regulating ferroptosis, with compromised mitochondrial morphology and functionalities primarily involved in this process. Various mitochondria-associated proteins and metabolic processes have been identified to modulate ferroptotic cell death.

Kaempferol Ameliorates the Inhibitory Activity of Dexamethasone in the Osteogenesis of MC3T3-E1 Cells by JNK and p38-MAPK Pathways

Baocheng Xie et al.

Summary: Kaempferol exhibits beneficial effects on osteogenic differentiation and counteracts the inhibitory activity of dexamethasone by activating JNK and p38-MAPK signaling pathways, improving bone mineralization. This potential makes it a promising candidate for developing new drugs for treating glucocorticoid-induced osteoporosis.

Summary: This study identified 68 differentially expressed genes mainly enriched in immune response and chemokine signaling pathway. Six hub genes were successfully identified with AUC>0.80 by ROC analysis and relative expression of P<0.05. RA synovial tissues had a higher proportion of 7 immune cells compared to OA, while 4 immune cells were found in relatively lower proportion (P<0.05).
