Summary: The Wnt pathway plays a crucial role in maintaining blood brain barrier (BBB) integrity and regulating cognitive decline in vascular cognitive impairment (VCI) and Alzheimer's disease (AD). The E3 ubiquitin ligase PDZRN3 is a key regulator of this pathway, impacting endothelial cell function and BBB permeability. Depletion of PDZRN3 in endothelial cells enhances BBB integrity and attenuates cognitive decline in both VCI and AD models. These findings suggest that targeting Wnt signaling could be a potential therapeutic strategy for preserving BBB function and cognitive health in neurodegenerative diseases.
