Immune cell profiling in atherosclerosis: role in research and precision medicine

Dawn M. Fernandez et al.

Summary: Inflammation plays a key role in atherosclerosis, and there is still a need to address cardiovascular risk in optimally treated patients. Evidence that targeting inflammation can reduce cardiovascular events highlights the urgent need for new immunotherapies tailored to specific patient immune alterations. Successful immunotherapies for atherosclerosis must be personalized based on distinct immune changes in patient groups.

Tutorial: guidelines for the computational analysis of single-cell RNA sequencing data

Tallulah S. Andrews et al.

Summary: Single-cell RNA sequencing (scRNA-seq) is a popular and powerful technology for profiling the whole transcriptome of individual cells, but analyzing the large volumes of data requires specialized statistical and computational methods. This article provides an overview of the computational workflow, common tasks and tools for addressing biological questions, as well as guidelines for best practices in computational analyses. It serves as a hands-on guide for experimentalists and an overview for bioinformaticians developing new computational methods.

miR-33 Silencing Reprograms the Immune Cell Landscape in Atherosclerotic Plaques

Milessa Silva Afonso et al.

Summary: Inhibition of miR-33 shifts the balance of proatherosclerotic and antiatherosclerotic immune cell subsets in atherosclerotic plaques, reducing plaque burden and inflammation. This treatment also affects the expression of specific target genes in different macrophage subsets, ultimately leading to a decrease in aortic CD8+ T cells and CD4+ T-helper 1 cells, and an increase in regulatory T cells within plaques. These findings shed light on the immune mechanisms underlying the atheroprotective effects of anti-miR-33.

Single cell analyses to understand the immune continuum in atherosclerosis

Christopher A. Hill et al.

Summary: The accumulation of lipids in the arterial wall triggers a network of inflammatory processes, contributing to the development of atherosclerosis. Recent clinical findings suggest that targeting specific immune alterations can prevent recurrent cardiovascular events. Single-cell approaches like CyTOF, scRNA-seq, and CITE-seq have emerged as valuable tools in understanding cellular plasticity within atherosclerotic lesions, but challenges remain in applying these methods to human vascular tissue.

Wnt signaling enhances macrophage responses to IL-4 and promotes resolution of atherosclerosis

Ada Weinstock et al.

Summary: IL-4 and IL-13 play crucial roles in the resolution of atherosclerosis inflammation, collaborating with other factors. Activation of Wnt signaling is inversely associated with disease severity in both mice and humans, enhancing macrophage responsiveness to IL-4 via a PGE(2)/STAT3 axis.
