Interaction with Ribosomal Proteins Accompanies Stress Induction of the Anticancer Metallodrug BOLD-100/KP1339 in the Endoplasmic Reticulum

Benjamin Neuditschko et al.

Summary: The study reveals that the anticancer agent BOLD-100 interacts with ribosomal proteins, induces ER stress, and modulates GRP78 in cancer cells. Target-profiling experiments and cellular observations provide evidence for these changes.

Defining the carrier proteome limit for single-cell proteomics

Tommy K. Cheung et al.

Summary: Single-cell proteomics by mass spectrometry (SCoPE-MS) is a novel method to quantify multiplexed single-cell proteomes, but it faces technical limitations related to isobaric labeling and mass spectrometry. Increasing carrier proteome levels requires a corresponding increase in the number of ions sampled to maintain quantitative accuracy. The introduction of SCPCompanion software assists in rapid evaluation of single-cell proteomic data and recommends parameters for improved data quality.

Spatiotemporal profiling of cytosolic signaling complexes in living cells by selective proximity proteomics

Mi Ke et al.

Summary: By developing more selective proximity labeling probes and combining them with label-free quantitative proteomics, the study enables exploration of cytosolic protein assemblies, such as phosphotyrosine-mediated protein complexes formed in response to minute-scale EGF stimulation. The authors demonstrate the application of this method in profiling the spatiotemporal interactome of the EGFR signaling component STS1.

Interdependence of metals and its binding proteins in Parkinson's disease for diagnosis

Athira Anirudhan et al.

Summary: Research on metalloproteins in Parkinson's disease patients reveals their crucial roles in environmental information processing, immune response, neuronal function, endocrine regulation, aging, and signal transduction pathways. Dysregulation of gene expression affects metal availability, while alterations in metal concentrations in serum and cerebrospinal fluid impact diagnosis of PD. This study suggests analyzing metal levels in serum and gene expression can provide a cost-effective diagnostic method for Parkinson's disease.

Comprehensive analysis of single cell ATAC-seq data with SnapATAC

Rongxin Fang et al.

Summary: The paper introduces SnapATAC, a software package for analyzing single cell ATAC-seq datasets, which can dissect cellular heterogeneity and map cellular states' trajectories. The Nystrom method allows processing data from up to a million cells, and it incorporates existing tools for single cell ATAC-seq dataset analysis. SnapATAC is applied to mouse secondary motor cortex profiles and identifies candidate regulatory elements and cell-type specific transcriptional regulators.

James H. Lovett et al.

Summary: X-ray absorption spectroscopy and X-ray fluorescence microscopy are nondestructive techniques that require minimal sample manipulation, often used to investigate the fate of metallodrugs in physiological samples. These methods are advantageous in studying metallodrug localization and speciation.

Cellular binding, uptake and biotransformation of silver nanoparticles in human T lymphocytes

Anzhela Malysheva et al.

Summary: The study demonstrates the presence of silver nanoparticles in nanoparticulate form after being taken up by human lymphocytes. Biotransformation of AgNPs is primarily dominated by sulfidation, serving as a detoxification pathway for cells. Toxicity of AgNPs is mainly driven by internalized silver, with differences in toxicity between nanoparticles of different coatings and sizes being minimized when expressed as intracellular mass of silver per cell.

Multi-target mode of action of silver against Staphylococcus aureus endows it with capability to combat antibiotic resistance

Haibo Wang et al.

Summary: This study identifies 38 authentic Ag+-binding proteins in Staphylococcus aureus and captures a molecular snapshot of the dynamic bactericidal action of Ag. It validates that Ag can inhibit a key target in S. aureus through binding to catalytic His185, leading to enhanced antimicrobial efficacy and resensitization of MRSA to antibiotics. The multi-target mode of action of Ag endows its sustainable antimicrobial efficacy and provides a potential approach for combating antimicrobial resistance.

Nano-scale imaging of dual stable isotope labeled oxaliplatin in human colon cancer cells reveals the nucleolus as a putative node for therapeutic effect

Anton A. Legin et al.

Summary: This study investigated the subcellular accumulation of oxaliplatin in colorectal cancer cells and found a pronounced tendency for cytoplasmic aggregation and effects on nuclear structures, particularly nucleoli. Interestingly, resistant cell lines showed disproportionately high accumulation of oxalate ligand, despite similar platinum distribution to sensitive cell lines.
