Phosphorescent iridium(III) complexes as lifetime-based biological sensors for photoluminescence lifetime imaging microscopy

Summary: Luminescent iridium(III) complexes have been extensively studied as intracellular sensors for photoluminescence microscopy imaging due to their high phosphorescence quantum yields, photostability, good cell membrane permeability, and rich and sensitive photophysical properties. Photoluminescence lifetime imaging microscopy (PLIM) allows for visualizing and mapping the decay of luminescent dyes in the time domain during cellular imaging. This review article focuses on the design and applications of lifetime-responsive phosphorescent iridium(III) complexes as imaging reagents for mapping environmental parameters and sensing analytes of interest in intracellular conditions.

Ferroptosis Photoinduced by New Cyclometalated Iridium(III) Complexes and Its Synergism with Apoptosis in Tumor Cell Inhibition

Summary: The limited therapeutic efficacy of photodynamic therapy (PDT) to hypoxic solid tumors has been improved by two new benzothiophenylisoquinoline-derived cyclometalated Ir-III complexes, IrL1 and MitoIrL2. Both complexes demonstrated a type I PDT process and induced photoinduced ferroptosis in tumor cells under hypoxia, with MitoIrL2 outperforming IrL1 in inhibiting tumor growth through a synergistic effect of ferroptosis and apoptosis.

Ruthenium(II) complexes coordinated to graphitic carbon nitride: Oxygen self-sufficient photosensitizers which produce multiple ROS for photodynamic therapy in hypoxia

Summary: The novel oxygen self-sufficient photosensitizer Ru-g-C3N4, designed to overcome tumor hypoxia-induced resistance in photodynamic therapy, shows enhanced water solubility, visible light absorption, and biocompatibility. Through catalyzing decomposition of H2O2 and H2O to generate O2, Ru-g-C3N4 significantly improves PDT efficacy by producing multiple reactive oxygen species and alleviating tumor hypoxia. This metal complex-grafted g-C3N4 photosensitizer represents a groundbreaking development in oxygen-producing photodynamic therapy agents.
