Cycloruthenated Self-Assembly with Metabolic Inhibition to Efficiently Overcome Multidrug Resistance in Cancers

Jia Li et al.

Summary: RuZ, a cycloruthenated complex, self-assembles into nanoaggregates in cell culture medium, leading to high intracellular concentrations in multi-drug resistant (MDR) cancer cells. It decreases oxygen consumption, inhibits glycolysis, lowers ATP levels, increases retention in MDR cells, induces oxidative stress and apoptosis. Proteomic analysis shows RuZ decreases glycolysis and mitochondrial respiration proteins while increasing apoptosis-related proteins. It inhibits proliferation of 35 cancer cell lines, including drug-resistant ones, and is effective in doxorubicin-resistant mouse tumor xenografts.

An ER-Targeting Iridium(III) Complex That Induces Immunogenic Cell Death in Non-Small-Cell Lung Cancer

Lili Wang et al.

Summary: The Ir1 iridium complex induces immunogenic cell death in non-small cell lung cancer cells, triggering an anti-tumor T-cell response and resulting in long-lasting anti-tumor immunity through the activation of ICD. Ir1 is the first Ir-based complex capable of developing an immunomodulatory response by inducing immunogenic cell death.

Regulating Tumor N6-Methyladenosine Methylation Landscape using Hypoxia-Modulating OsSx Nanoparticles

Yue Zheng et al.

Summary: A new type of nanocatalyst nanoparticles has been developed to downregulate genes involved in hypoxia pathway, elevate RNA m(6)A methylation levels, and degrade mRNA of hypoxia-related genes, thereby improving tumor hypoxia. This research sheds light on the potential of regulating RNA modification to overcome chemoresistance, providing a new avenue for understanding the underlying mechanisms of hypoxia-modulating nanomaterials.

Using bio-orthogonally catalyzed lethality strategy to generate mitochondria-targeting anti-tumor metallodrugs in vitro and in vivo

Xuling Xue et al.

Summary: Through a bio-orthogonally catalyzed lethality (BCL) strategy, a targeting anti-tumor metallodrug Ru-rhein was successfully generated and demonstrated significant anti-tumor efficacy in vitro and in vivo. This strategy could also be extended to other metal complexes for potential cancer treatment applications.

Ferroptosis Photoinduced by New Cyclometalated Iridium(III) Complexes and Its Synergism with Apoptosis in Tumor Cell Inhibition

Hao Yuan et al.

Summary: The limited therapeutic efficacy of photodynamic therapy (PDT) to hypoxic solid tumors has been improved by two new benzothiophenylisoquinoline-derived cyclometalated Ir-III complexes, IrL1 and MitoIrL2. Both complexes demonstrated a type I PDT process and induced photoinduced ferroptosis in tumor cells under hypoxia, with MitoIrL2 outperforming IrL1 in inhibiting tumor growth through a synergistic effect of ferroptosis and apoptosis.
