A tumor microenvironment responsive nanoplatform with oxidative stress amplification for effective MRI-based visual tumor ferroptosis

Shiwei Luo et al.

Summary: Ferroptosis is a promising non-apoptotic regulated cell death form for cancer treatment. However, the efficacy of ferroptosis is limited by high glutathione levels and insufficient hydrogen peroxide levels in tumors. In this study, a theranostic nanoplatform was designed to enhance ferroptosis in the tumor microenvironment by releasing active components, and it was also used for tumor-specific magnetic resonance imaging, exhibiting high specificity and efficacy.

Broadening horizons: the role of ferroptosis in cancer

Xin Chen et al.

Summary: Ferroptosis is an iron-dependent form of regulated cell death driven by excessive lipid peroxidation, and can be induced by various agents to suppress tumor growth while also potentially triggering inflammation-associated immunosuppression. The extent of ferroptosis's impact on tumor biology and its interactions with other signaling pathways are still under investigation.

Generation of hydroxyl radical-activatable ratiometric near-infrared bimodal probes for early monitoring of tumor response to therapy

Luyan Wu et al.

Summary: The response of tumors to radiotherapy or ferroptosis is closely linked to hydroxyl radical production, and accurate imaging of this reactive oxygen species may allow for monitoring of therapy response. A ratiometric probe has been developed for in vivo imaging of hydroxyl radical generation, providing a reliable method for monitoring treatment response in tumors undergoing erastin-induced ferroptosis or radiotherapy.

Metabolic Control by Heat Stress Determining Cell Fate to Ferroptosis for Effective Cancer Therapy

Shaowei Xie et al.

Summary: The flexible manipulation of cancer cell fate through exogenous stimulation-induced metabolic reprogramming can effectively handle therapy resistance and sensitize tumors to ferroptosis. The proposed heat-triggered tumor-specific ferroptosis strategy utilizing nanoformulation showed promising antitumor effects by inducing oxidative damage and targeting key pro-ferroptotic factors. This work provides insights and strategies for heat-triggered ferroptosis in situ, offering a potential alternative to direct ferroptosis inducers with reduced side effects.
