Data-Independent Acquisition for the Detection of Mononucleoside RNA Modifications by Mass Spectrometry

Summary: RNA is modified in cells to modulate molecular functions and processes. Over 140 modifications have been identified, with tRNA having the highest density and diversity of modifications. Next-generation sequencing (NGS) and mass spectrometry (MS) are used to identify and quantify these modifications. Mass spectrometry can accurately identify multiple modifications in a single run. Studies have found that multiple tRNA wobble position modifications are upregulated in salt-stressed growth conditions in Arabidopsis thaliana, potentially stabilizing translation of salt stress induced proteins.
