Impact of donation after circulatory death donor allografts on outcomes following liver transplantation for fulminant hepatic failure in the United States

Summary: Limited data exist on the impact of DCD allografts on outcomes following liver transplantation in patients with fulminant hepatic failure. Graft and patient survival in DCD-FHF were found to be comparable to DBD-FHF and DCD-non-FHF over a 15-year period, with improvements observed in long-term survival. Recipient age, male gender, African American ethnicity, donor age, and cold ischemia time were identified as predictors of survival in FHF, while DCD status was only predictive of graft survival. Consideration of DCD in FHF could help expand the donor pool for critically ill patients.

Scaled preparation of extracellular vesicles from conditioned media

The scalability of conventional EV preparation methods poses a major challenge in the therapeutic EV field, requiring the identification of appropriate concentration and purification methods for scale-up. Adopting principles from enveloped viruses, which have been used in clinics for over two decades, can provide valuable insights for EV manufacturing.

Effective methods for isolation and purification of extracellular vesicles from plants

This study established and verified quartz crystal microbalance (QCM) as a highly sensitive label-free immunosensing technique for characterizing clinically approved umbilical cord MSC-EVs. By using QCM along with common characterization methods, specific detection of EVs via EV and MSC markers was achieved, allowing for quantitative determination of the ratio of marker-specific EVs versus non-vesicular particles.

Researchers found that tomato fruit is a good source for obtaining NVs. During the isolation process of NVs from tomatoes, co-isolation of viral particles was occasionally observed. Density gradient ultracentrifugation was effective in separating NVs from viral particles, and cryo-TEM, SEM, and proteomics were useful in detecting possible viral contamination.

Furthermore, PJ-EV-treated DCs have shown strong potential in inducing the proliferation and differentiation of T cells towards Th1-type and cytotoxic T cells.
