Modulation of designer biomimetic matrices for optimized differentiated intestinal epithelial cultures

Wang Xi et al.

Summary: This study developed hydrogel scaffolds with different elasticities and investigated their influence on the expansion, organization, and differentiation of intestinal epithelial cells. The results showed that cells on soft substrates exhibited improved characteristics resembling in vivo intestinal tissue, which is of significant importance for the design of biomaterials for ex vivo intestinal models.

Organoid engineering with microfluidics and biomaterials for liver, lung disease, and cancer modeling

Su Kyeom Kim et al.

Summary: Advancements in organoid technology have allowed for better replication of in vivo organ environments, overcoming limitations of existing disease models. Integration of engineering approaches such as microfluidics and biomaterials is necessary for more precise disease modeling. Combined with organoids, these engineered disease models provide a powerful platform for personalized medicine research.

SARS-CoV-2 Infection and Disease Modelling Using Stem Cell Technology and Organoids

Marta Trevisan et al.

Summary: This Review briefly describes the virology and pathogenesis of SARS-CoV-2, emphasizing how stem cell technology and organoids can help understand the cell tropisms of SARS-CoV-2 and its disease mechanisms in the human host. Studies using these technologies showed that lung epithelial cells, gastrointestinal cells, heart cells, and choroid plexus epithelial cells are key targets of SARS-CoV-2, leading to cell damage and inflammation. Neurological symptoms and vascular damage are suggested to be indirect effects of systemic inflammatory and immunological responses to SARS-CoV-2 infection, as endothelial cells and neurons are less susceptible to viral infection.

To form and function: on the role of basement membrane mechanics in tissue development, homeostasis and disease

Nargess Khalilgharibi et al.

Summary: The basement membrane plays a crucial role in tissue development and diseases, with its complex mechanical properties changing through different molecular mechanisms, thereby affecting tissue form and function. Investigating the mechanical properties of the basement membrane is important for disease diagnosis and treatment, as well as the development of better in silico and in vitro models, which can also reduce the use of animals in research.

Angiotensin-Converting Enzyme 2 (ACE2) as a Potential Diagnostic and Prognostic Biomarker for Chronic Inflammatory Lung Diseases

Dejan Marcetic et al.

Summary: Chronic inflammatory lung diseases are characterized by uncontrolled immune response in the airways, and the lack of specific diagnostic and therapeutic biomarkers poses a major challenge for pulmonologists. Approved therapeutic approaches focus on achieving disease remission, while the protective role of ACE2 in the lungs and its exact pathophysiological mechanism of action remain unclear.

Summary: Organ-on-a-Chip (OOAC) systems are engineered microfluidic systems that mimic the physiological environment of specific organs and have the potential to replace animal models. The success of cell cultures within these systems depends on the choice of scaffold, highlighting the critical role of scaffolds in OOAC research.
