Novel oxindole compounds inhibit the aggregation of amyloidogenic proteins associated with neurodegenerative diseases

Identification of novel neuroprotective N,N-dimethylaniline derivatives that prevent oxytosis/ferroptosis and localize to late endosomes and lysosomes

Summary: The study identified nine neuroprotective compounds, with N,N-dimethylaniline as a common structure, that effectively prevent oxytosis/ferroptosis by reducing reactive oxygen production and decreasing ferrous ions. The most potent derivatives, GIF-2114 and GIF-2197-r, target late endosomes/lysosomes and protect hippocampal cells from oxidative stress and neurodegenerative disorders by inhibiting oxytosis/ferroptosis. These compounds offer a new therapeutic approach for neurodegenerative diseases.
