Proteolysis-targeting chimera against BCL-XL destroys tumor-infiltrating regulatory T cells

Ryan Kolb et al.

Summary: The pharmacological degradation of BCL-X-L preferentially induces apoptosis of tumor-infiltrating Tregs, promoting CD8 T cell activation and effective suppression of tumor growth without causing damage to normal tissues or thrombocytopenia. This finding suggests that targeting BCL-X-L could be a potential therapeutic strategy for cancer immunotherapy.

Development of Triantennary N-Acetylgalactosamine Conjugates as Degraders for Extracellular Proteins

Yaxian Zhou et al.

Summary: Targeted protein degradation (TPD) technology has become a focus of attention for researchers due to its potential as a new therapeutic modality and selective removal of various protein targets. Specifically, LYsosome TArgeting Chimera (LYTAC) has emerged as a promising technology for degrading extracellular protein targets, using the cation-independent mannose-6-phosphate receptor (CI-M6PR) to deliver them to lysosomes. This study successfully utilized the asialoglycoprotein receptor (ASGPR) expressed on liver cells, with the ligand triantennary N-acetylgalactosamine (tri-GalNAc), to target extracellular proteins for degradation, demonstrating cell type specificity in liver cell lines.

Chemoenzymatic glycan-selective remodeling of a therapeutic lysosomal enzyme with high-affinity M6P-glycan ligands. Enzyme substrate specificity is the name of the game

Xiao Zhang et al.

Summary: Functionalization of therapeutic lysosomal enzymes with M6P glycan ligands was achieved through chemical synthesis of truncated M6P-glycan oxazolines, allowing for enzymatic glycan remodeling of rhGAA. This method significantly improved the binding affinity for the CI-MPR receptor, leading to enhanced therapeutic efficacy in an in vitro cell model of Pompe disease. This study provides a promising strategy for enhancing the effectiveness of enzyme replacement therapy through site-selective M6P-glycan remodeling of recombinant lysosomal enzymes.

Degradation from the outside in: Targeting extracellular and membrane proteins for degradation through the endolysosomal pathway

Green Ahn et al.

Summary: Targeted protein degradation (TPD) is a promising strategy to remove deleterious proteins for therapeutic benefit and to probe biological pathways. Various technologies have been developed to degrade challenging cytosolic targets, but there is a need for approaches to degrade extracellular and membrane proteins. Recent advancements in technology, such as lysosome-targeting chimeras (LYTACs), show potential in degrading both secreted and membrane-anchored targets.
