Inhibition of EZH2 primes the cardiac gene activation via removal of epigenetic repression during human direct cardiac reprogramming

Yawen Tang et al.

Summary: Cardiovascular disease remains the leading cause of death in the United States due to the limited regenerative capacity of adult hearts. Inhibition of EZH2, an important epigenetic barrier, significantly increases the efficiency of converting non-myocytes into induced cardiomyocytes, promoting activation of cardiac genes and supression of collagen genes. This inhibition facilitates the activation of cardiac genes in fibroblasts and eases the production of hiCMs.

A Minimally Invasive Exosome Spray Repairs Heart after Myocardial Infarction

Jialu Yao et al.

Summary: Myocardial infarction is the most common cause of death worldwide, with heart failure becoming a major determinant of adverse prognosis in survivors. Exosome spray (EXOS) based on MSC exosomes and biomaterials shows promise in improving heart function, reducing fibrosis, and promoting angiomyogenesis for heart repair, making it a feasible and safe strategy for future therapeutic delivery.

The cardioprotective effects of hydrogen sulfide by targeting endoplasmic reticulum stress and the Nrf2 signaling pathway: A review

Fatemeh Yarmohammadi et al.

Summary: Cardiac diseases are on the rise due to lifestyle changes, urbanization, and aging, with oxidative stress playing a key role in cardiac injury progression. Hydrogen sulfide (H2S) has shown potential in improving various cardiac conditions through antioxidant and anti-ER stress-related activities, by regulating Nrf2 expression and its signaling pathway in myocytes.

Platelet membrane and stem cell exosome hybrids enhance cellular uptake and targeting to heart injury

Shiqi Hu et al.

Summary: In this study, stem cell-derived exosomes were hybridized with platelet membranes to enhance their targeting ability for injured hearts and reduce uptake by macrophages. The hybridization induced macropinocytosis, significantly increasing cellular uptake by endothelial cells and cardiomyocytes. In vivo studies demonstrated the cardiac targeting ability of these hybrid exosomes in a mouse model of myocardial infarction, suggesting a potential therapeutic benefit over non-modified exosomes.
