Alzheimer's Disease Genetics: A Dampened Microglial Response?

Zena K. Chatila et al.

Summary: Alzheimer's disease is an age-related neurodegenerative condition, and microglia cells have been found to play a central role in its pathogenesis. Genetic studies have identified dysfunctional microglial pathways associated with genetic risk factors for AD. These findings provide potential therapeutic targets for intervention.

Functional insights from biophysical study of TREM2 interactions with apoE and Aβ1-42

Daniel L. Kober et al.

Summary: The study using comprehensive biolayer interferometry analysis revealed that TREM2 binds apoE through a protein-mediated mechanism, with slight affinity differences among isoforms; TREM2 and sTREM2 can directly bind mAβ42 and potently inhibit Aβ42 polymerization, suggesting a potential role in preventing AD pathogenesis.

CD33-Targeted Therapies: Beating the Disease or Beaten to Death?

Joseph E. Maakaron et al.

Summary: CD33 is a transmembrane protein that is highly expressed on AML progenitor cells, making it a specific target for AML therapeutics. Various targeted therapies, including gemtuzumab ozogamicin, have been developed, but more precise strategies such as cellular therapy mechanisms and linker molecules are currently being explored for clinical benefit.

Prior activation state shapes the microglia response to antihuman TREM2 in a mouse model of Alzheimer's disease

Daniel C. Ellwanger et al.

Summary: TREM2 plays a critical role in microglia activation trajectories, with ligand engagement being essential for certain activation pathways. Activation trajectories induced by stimuli are more prominent in female mice than male mice. Injection of hT2AB can replenish depleted microglial pools lacking certain activation trajectories.

The role of innate immune genes in Alzheimer's disease

Ana Griciuc et al.

Summary: Recent studies have highlighted the crucial role of the innate immune system and neuroinflammation in the pathogenesis of Alzheimer's disease, with a focus on microglial receptors CD33 and TREM2. Therapeutic approaches targeting CD33 and TREM2, such as gene therapy, small molecules, and immunotherapy, have shown promise in modulating neuroinflammation and potentially reducing neurodegeneration in Alzheimer's disease. Antibodies against CD33 and TREM2 have entered clinical trials and may provide a new avenue for treating Alzheimer's disease by targeting neuroinflammation.

Development of a robust crystallization platform for immune receptor TREM2 using a crystallization chaperone strategy

Noel J. Byrne et al.

Summary: In this study, a robust crystallization platform was developed for the TREM2 Ig-like domain by fusing it with the structural chaperone MBP. The produced protein crystals diffracted at 2.0 angstrom, indicating suitability for potential ligand soaking. Crystal packing analysis showed that most of the surface of the TREM2 Ig domain is available for small molecule binding, and a proof of concept co-crystallization study validated the utility of this system for the discovery of new TREM2 therapeutics.
