Biofabricating murine and human myo-substitutes for rapid volumetric muscle loss restoration

Marco Costantini et al.

Summary: This study proposes a biofabrication approach to restore skeletal muscle mass, tissue structure, and functionality rapidly, efficiently guiding cell differentiation and myobundle formation by replicating muscle anisotropic organization. Upon implantation, the biofabricated muscle substitutes support the formation of new blood vessels and neuromuscular junctions, contributing to muscle mass and force recovery. These findings provide a strong basis for further testing in large animal models and potential translation into clinical settings.

Microfluidic encapsulation of nanoparticles in alginate microgels gelled via competitive ligand exchange crosslinking

Victor Dal Posolo Cinel et al.

Summary: Efficient delivery of nanometric vectors complexed with nanoparticles to target tissues is crucial for gene therapy, and one possible solution is to confine these vectors within microgels for slow and local release. Alginate microgels with high encapsulation efficiency and continuous release of nanoparticles over time are considered valuable for gene therapy research.

Probing Single-Cell Macrophage Polarization and Heterogeneity Using Thermo-Reversible Hydrogels in Droplet-Based Microfluidics

B. M. Tiemeijer et al.

Summary: To study the biological function of adherent cell types at single-cell resolution with high throughput, combining droplet-based microfluidics with hydrogels is a versatile and powerful tool. It allows for robust droplet formation at low temperatures, catalyzer-free droplet gelation, and easy cell recovery after culture for downstream analysis.
